Apoptose e um tipo de morte celular geneticamente regulada, com características morfológicas e bioquímicas específicas, essencial na organogênese e na manutenção da homeostase do organismo. Este evento celular tambem está envolvido na patogênese de diferentes doenças. Nas doenças parasitárias, como na doença de Chagas, tem sido demonstrada a importancia deste evento para o estabelecimento da infecção pelo Trypanosoma cruzi. Na leishmaniose, poucos trabalhos tem investigado a participação da apoptose nos mecanismos de resistencia e susceptibilidade desta doença. Nós avaliamos a ocorrencia deste evento no modelo de leishmaniose murino, utilizando camundongos isogênicos CBA infectados com L. amazonensis, para as quais são susceptíveis, ou L. major, para as quais são resistentes. Diferente de outros modelos que utilizam camundongos com background genético distintos, este modele permite apontar para fatores relacionados ao parasito que possam estar determinando o padrão de resposta. Foram utilizadas diferentes técnicas para a detecção da apoptose em diferentes fases deste processo. Células do linfonodo de drenagem foram avaliadas no intervalo de 7 a 42 dias de infecção. Nós observamos através da contagem com a coloração de azul de tripan, que houve um aumento no número de células mortas nos linfonodos dos animais infectados com L. amazonensis, porém em relação ao número de células totais, o percentual de células mortas foi semelhante entre os dois grupos. Através da análise na citometria de fluxo com marcação com anexina-V, verificou-se baixa frequência de celulas apoptóticas, tanto em celulas totais como em linfocitos T CD4+ e TCD8+. Porem, verificou-se uma diminuição na frequencia de celulas CD4+ nos animais infectados. Na analise in situ atraves da marcação com a técnica de TUNEL, observou-se com sete dias um maior numero de células apoptóticas nos animais infectados com L.major e com 21 dias um maior numero de celulas em apoptose nos animais infectados com L. amazonensis, no 42° dia nao houve diferenças entre os grupos. Na microscopia eletrônica foram evidenciados os aspectos típicos de apoptose nos dois grupos, sendo que foram observados com maior frequencia macrófagos com corpos apoptoticos nos linfonodos dos animais infectados com L. amazonensis no 42° dia. A analise histopatologica mostra aumento no numero de macrofagos com 42 ° dia nos animais infectados com L. amazonensis. Os dados apresentados mostram diferenças na cinetica e na frequencia da ocorrencia de apoptose na resposta de resistencia e de susceptibilidade.
Abstract
Apoptosis Is a type of genetically regulated cell death, with specific morphological and biochemical characteristics, and it is essential in organogenesis and maintenance of homeostasis of the organism. This cellular event is also involved in the pathogenesis of different diseases. In parasite- caused diseases, such as Chagas' disease, the importance of this event for the establishment of infection by Trypanosoma cruzi has been demonstrated. In leishmaniasis, few studies have investigated the participation of apoptosis in the mechanism of resistance and susceptibility in this disease. We evaluated the occurrence of this event in the murine leishmaniasis model, using isogenic CBA mice infected with L amazonensis, to which they are susceptible, or L major, to which they are resistant. Different from the other models that use mice with distinct genetic background, this model allows the identification of factors related to the parasite which could be determining the response pattern. Different techniques were used for the detection of apoptosis in different phases of this process. Cells from the draining lymph node were evaluated in intervals of 7 to 42 days of infection. We observed, through counting with tripan blue coloring, that there was an increase in the number of killed cells in the lymph nodes of the animals infected with L amazonensis, however, in relation to the total number of cells, the percentage of killed cells was similar between both groups.Through the analysis of flow cytometric staining with annexin-V, a low frequency of apoptotic cells, both in total cells and in T CD4+ and CD8+ lymphocytes was detected. However, a decrease in the frequency of CD4+ cells in the infected animals was detected. The in situ analysis through staning with TUNEL technique displayed a higher number of apoptotic cells in the infected animals with L.major for 7 days, and at 21 days a higher number of cells in apoptosis in the animals infected with L amazonensis, on the 42"*^ day there was no difference between the groups. In electronic microscopy the typical aspects of apoptosis were evidenced in both groups, but macrophages were with apoptotic bodies in the lymph nodes were observed with more frequency in the animals infected with L. amazonensis on the 42"*^ day. The histopathologic analysis showed an increase in the number of macrophages on the 42"^^ day in the animals infected with L. amazonensis. The data presented show differences in kinetics and frequency of occurrence of apoptosis in resistance, as well as susceptibility.
