Neutrófilos são componentes importantes do sistema imune e fazem parte da primeira linha de defesa contra infecções. Na infecção intradérmica com L. braziliensis demonstramos que os neutrófilos são constantemente recrutados ao sítio de infecção. Além disso, já foi demonstrado que a interação de neutrófilos com macrófagos regula a resposta contra L. major. Neste estudo, investigamos o papel dos neutrófilos na infecção causada por Leishmania braziliensis usando o modelo de infecção intradérmica. Neutrófilos foram purificados de camundongos BALB/c, após a injeção intraperitonial de tioglicolato, sua pureza foi avaliada pela marcação com Gr-1, utilizando a citometria de fluxo. A posterior co-cultura de neutrófilos vivos ou mortos pelo calor com macrófagos peritoniais infectados com L. braziliensis levou a uma redução significativa na taxa de infecção e no número de amastigotas por célula quando comparado com as culturas controle. A presença de neutrófilos vivo ou mortos na cultura de macrófagos infectados com L. braziliensis foi associada a produção de altos níveis de TNF-a. Para avaliar o papel dos neutrófilos in vivo, camundongos foram co-inoculados com neutrófilos vivos e L. braziliensis. Observou-se que estes animais desenvolveram lesões significativamente menores e apresentaram um número menor de parasitas quando comparados com os camundongos controle. Por último, camundongos BALB/c foram depletados de neutrófilos após injeção intraperitonial do anticorpo RB6-8C5 e foram, em seguida, infectados com L. braziliensis. Camundongos depletados apresentaram lesões e carga parasitária maiores quando comparados com camundongos controle, os quais receberam injeção de IgG de rato. Estes dados indicam que os neutrófilos são células fundamentais para a eliminação inicial de L. braziliensis em camundongos BALB/c.

Neutrophils are important components of the immune system and provide the first mechanism of defense against infection. In the dermal experimental model of infection with L. braziliensis, we observed that neutrophils are constantly recruited to the lesion site, throughout the infection period. Moreover, it was observed that the interaction between macrophages and neutrophils regulates infection with L. major. In the present work we investigated the role of neutrophils in the infection caused by Leishmania braziliensis using a mouse intradermal model of infection. Neutrophils were obtained from mouse peritoneal cavities by injection of thioglycolate broth, purity was assesses by flow cytometry using Gr-1 staining. Live and heat-killed neutrophils incubation with L. braziliensis infected peritonial macrophages from BALB/c mice led to a significant decrease in the infection rate as well as in the number of amastigotes per cell when compared to cells infected with promastigotes only. Co-incubation of live or heat-killed neutrophils with macrophages infected with L. braziliensis was associated with the production of high leveis of TNF-a. To investigate the role of neutrophils in the in vivo infection, BALB/c mice were co-inoculated with L. braziliensis and neutrophils. In these experiments, we observed that co-inoculation of live neutrophils and L. braziliensis in the ear dermis led to the development of smaller lesions as well as a decrease in parasite load. On the contrary, when mice were depleted of neutrophils by injection of RB6- 8C5 antibody, we observed that depleted animais showed an increase in lesion size and in parasite load when compared to animais injected with control rat IgG antibody. According to our data, neutrophils are essencial in the inicial elimination of L braziliensis in BALB/c mice.
