POTENCIAL ANTI-HIPERTENSIVO DE LINALOL COMPLEXADO À BETA-CICLODEXTRINA: ESTUDOS DE FARMACODINÂMICA E FARMACOCINÉTICA

INTRODUÇÃO: A hipertensão arterial (HA) é importante problema de saúde pública e apresenta difícil controle e tratamento. Alternativas terapêuticas precisam ser encontradas e a pesquisa com produtos naturais é bastante promissora. Os óleos essenciais apresentam efeitos biológicos sobre o sistema cardiovascular e o monoterpeno linalol (LIN) tem sido estudado como anti-hipertensivo. Os monoterpenos apresentam dificuldades de solubilidade devido às suas características lipofílicas e a formação de complexos de inclusão principalmente com ciclodextrinas (CDs) demonstram ser de fácil síntese e utilização para estabilização e melhoria na farmacocinética e farmacodinâmica. OBJETIVO: avaliar a ação de linalol puro e do complexo de inclusão contendo linalol em β-ciclodextrina (LIN/β-CD) sobre os parâmetros cardiovasculares de pressão arterial (PA) e frequência cardíaca (FC) de ratos espontaneamente hipertensos (SHR) e normotensos Wistar. MATERIAL E MÉTODOS: Protocolos realizados de acordo à literatura e sinalizados na próxima sessão. RESULTADOS: Nos estudos de farmacocinética, o complexo LIN/β-CD foi capaz de aumentar a absorção, a biodisponibilidade de linalol na corrente sanguínea em cerca 20 vezes, assim como houve aumento da área sobre a curva de concentração sanguínea em relação ao tempo, demonstrando que uma vez complexado, LIN permanece por muito mais tempo na corrente sanguínea. Nos estudos para medida direta de PA e FC, administrações i.v. de LIN e LIN/β-CD (50 mg/kg) demonstraram ser capazes de induzir hipotensão em ratos SHR. Após administração oral, foi possível averiguar hipotensão e bradicardia por parte do grupo LIN/β-CD (50 mg/kg) quando comparado ao LIN especialmente nas 3 últimas horas. A administração de LIN e LIN/β-CD por via orogástrica de maneira subcrônica durante 21 dias (50mg/kg) em SHR induziu manutenção dos níveis pressóricos iniciais e apresentou um efeito anti-hipertensivo superior ao LIN e estatisticamente significante a partir do 15º dia, perdurando até o 20º dia, já o grupo LIN alcançou diminuição da pressão arterial apenas no último dia do tratamento. Adicionalmente, LIN/β-CD foi capaz de promover aumento nos níveis da citocina anti-inflamatória IL-10; foi capaz também de impedir o desenvolvimento de hipertrofia, com diminuição significante do índice de massa do coração, quando comparado ao grupo tratado com LIN. Em estudos de reatividade vascular com artéria mesentérica superior, houve vasorrelaxamento induzido por NPS (10-13-10-5M) sem alterações significantes em relação aos controles. A curva em resposta à Phe (10-10-10-6M) apresentou uma diminuição significante da potência farmacológica do grupo LIN/β-CD em relação ao grupo veículo. Com relação ao tratamento crônico de 60 dias o grupo LIN/β-CD apresentou efeito anti-hipertensivo importante, desde 15 dias de tratamento, perdurando durante todo o tratamento de 60 dias, aproximando-se dos animais normotensos sadios. O grupo LIN apresentou, em menor proporção, efeito anti-hipertensivo aproximando-se do grupo veículo. Com relação aos estudos de hipertrofia do coração, após 60 dias de tratamento, apenas o grupo tratado com o complexo LIN/β-CD apresentaram diminuição do índice de massa cardíaca quando comparado ao grupo veículo. Em estudos de reatividade vascular, após 60 dias de tratamento a curva concentração resposta à Phe apresentou efeito reduzido com deslocamento da curva para esquerda, sendo estatisticamente significante no grupo tratado com LIN/β-CD, quando comparado ao grupo veículo, sendo que o grupo linalol também apresentou esta diminuição da reatividade vascular, porém em menor proporção. Nas curvas de relaxamento, foi possível observar alterações estatisticamente significantes para o relaxamento induzido por Ach e NPS com relação ao grupo tratado com LIN LIN/CD em comparação as artérias de animais hipertensos tratados com veículo. Com relação aos estudos morfológicos, a histologia demonstrou que os grupos tratados com LIN e LIN/β-CD apresentaram características semelhantes ao grupo wistar sadio, sem sinais de injúrias. CONCLUSÃO: Os resultados em conjunto, sugerem que o complexo de inclusão LIN/β-CD tem promissor potencial de aumento da biodisponibilidade de LIN e melhora dos seus efeitos sobre o sistema cardiovascular com efeito hipotensor e anti-hipertensivo.

Abstracts:INTRODUCTION: Arterial hypertension (AH) is an important public health problem and its control and treatment have been increasingly difficult. Therapeutic alternatives need to be found and the search for new drugs with natural products is very promising. Essential oils have biological effects on the cardiovascular system and monoterpene linalool (LIN) has been studied as an antihypertensive. Despite the associated biological effects, monoterpenes have difficulties in solubility due to their lipophilic characteristics. These problems can be overcome with the formation of inclusion complexes with cyclodextrins (CDs). There is stabilization and improvements in the pharmacokinetics and pharmacodynamics of these unstable substances. AIM: to evaluate the action of pure linalool and the inclusion complex containing linalool in β-cyclodextrin (LIN/β-CD) on the cardiovascular parameters of blood pressure (BP) and heart rate (HR) of spontaneously hypertensive rats (SHR) and normotensive wistar. METHODS: All the scientific protocols onwards. RESULTS: Pharmacokinetic studies, LIN/β-CD was able to increase the bioavailability of linalool in the bloodstream by about 20 times, an increase in the area on the blood concentration curve in relation to time, demonstrating that a Once complexed, LIN remains in the bloodstream much longer; the excretion and elimination of complexed linalool was also lower when compared to LIN, data obtained through the clearance of substances. In studies for direct measurement of BP and HR, intravenous administrations of LIN and LIN/β-CD (50 mg/kg) have been shown to be able to induce hypotension in SHR This effect was associated with bradycardia when applied to pure linalool and tachycardia without statistical differences when applied to the inclusion complex and compared to the vehicle group. After oral administration and observation of direct effects for 6 hours, it was possible to ascertain hypotension and bradycardia by the LIN/β-CD group (50 mg/kg) when compared to LIN especially in the last 3 hours. The administration of LIN and LIN/β-CD via orogastric subchronic for 21 days in SHR induced maintenance of initial blood pressure levels and had an antihypertensive effect superior to LIN and statistically significant from the 15th day lasting until the 20th day, the vehicle group showed a significant increase until the end of the treatment; the LIN group achieved a decrease in blood pressure only on the last day of treatment. In addition, complexed linalool was able to promote an increase in the levels of the anti-inflammatory cytokine IL-10. The treatment with LIN/β-CD was able to prevent the development of cardiac hypertrophy, with a significant decrease in the heart mass index, when compared to the group LIN. In studies of vascular reactivity with superior mesenteric artery, there was vasorelaxation induced by SNP (10-13 -10-5M) without significant changes in relation to controls. The curve in response to PHE (10-10-10-6M) showed a significant decrease in the pharmacological potency of the linalool group in relation to the vehicle group. Regarding the chronic treatment of 60 days, the LIN/β-CD group had an important antihypertensive effect, since 15 days of treatment, lasting throughout the 60-day treatment, approaching healthy normotensive animals. The LIN group had, to a lesser extent, an antihypertensive effect approaching the vehicle. Regarding studies of heart hypertrophy, after 60 days of treatment, only the group treated with the LIN/β-CD complex and the wistar animals showed a decrease in the cardiac mass index when compared to the vehicle SHR group. In vascular reactivity, 60 days of treatment concentration curve response to Phe showed a reduced effect with shift of the curve to the left statistically significant in LIN/β-CD group, when compared to vehicle, and the LIN also presented this decrea reactivity, but to a lesser extent. In the relaxation curves, it was possible to observe statistically significant changes for the relaxation induced by ACH and SNP in relation to the group treated with LIN/β-CD compared to the vehicle. Regarding morphological studies, histology has shown that the groups treated with LIN and LIN/β-CD showed similar characteristics to the healthy Wistar group, with no signs of injuries. CONCLUSION: These results together suggest that the LIN/β-CD inclusion complex has a promising potential for increasing the bioavailability of LIN and improving its effects on the cardiovascular system.
