O metabólito 1-hidroxipireno foi determinado em amostras biológicas (urina) em
trabalhadores do CESTEH/FIOCRUZ, fumantes e não fumantes.
As amostras foram coletadas no final do expediente, no período de fevereiro a
março de 2001, sofreram hidrólise enzimática com a enzima β-Dglucoronidase/arilsulfatase (100.000 Fishman U/mL e 800.000 Roy U/mL), limpeza
(clean-up) por extração em fase sólida (Sep – Pak Vac 6cc (1g) C-18) e foram
analisadas por CLAE com detector de fluorescência.
Concentrações de 1-OHP entre 0,104 e 0,289 µg/g de creatinina foram
encontradas em amostras de fumantes. Perfis bem abaixo foram encontradas em não
fumantes, entre 0,002 e 0,020µg/g de creatinina.
A presença de 1-OHP em algumas amostras de não fumantes foi atribuída a
exposição ambiental, ativação metabólica, suscetibilidade individual, estado de
saúde, nutrição, idade, variações fisiológicas provavelmente do tecido adiposo do
corpo, intensidade de inspirar, o pH da urina, variações genéticas e atividade
enzimática.
Amostras de urina de fumantes que não apresentaram níveis de 1-OHP, foi
atribuída a fatores específicos individuais como variações fisiológicas, intensidade de
inspirar, o pH da urina, variações genéticas e atividade enzimática, suscetibilidade
individual e outros fatores , tais como gênero, etnia, idade, estado de saúde, nutrição e
poliformismo genético.
A identificação e quantificação de 1-OHP em fumantes e não fumantes são
inéditos no Brasil.
Resumen en ingles
The metabólito 1-hidroxipireno was determined in biological samples
(urine) in workers of CESTEH/FIOCRUZ, smokers and non smokers.
The samples were collected in the end of the file, from February to March of
2001. Samples were subnitted to enzymatic hydrolisys (β-Dglucoronidase/arylsulfatase 100,000 Fishman U/mL and 800,000 Roy U/mL), and
cleaning (clean-up) by extraction solid phase (Sep – Pak Vac 6cc C-18 ). After
that, samples were analyzed by CLAE with fluorescencie detector.
Concentrations of 1-OHP between 0,104 and 0,289 µg/g creatinine were
found in smokers' samples. Quite lower concentrations were found in non
smokers, between 0,002 and 0,020µg/g of creatinine.
The presence of 1-OHP in some “non smokers” samples and its
absence inothers “smokers” samples can be explained by the environmental
exposition, metabolic activation, individual susceptibility, health condition,
nutrition, age, physiologic variations probably of the fabric fatty of the body,
intensity of inspiring, the pH of the urine, genetic variations and enzymatic
activity.
The identification and quantification of 1-OHP in smokers and non
smokers are unpublished in Brazil.
