Metalotioneínas (MTs) são proteínas citosólicas cuja função biológica
está relacionada à regulação de metais essenciais e a detoxificação de metais
tóxicos. O conhecimento da compartimentalização intracelular de metais, da
função fisiológica das MTs e dos fatores que controlam seus níveis em
diferentes espécies de peixes é essencial para compreensão dos efeitos tóxicos
induzidos por metais. Geophagus brasiliensis (acará) é uma espécie nativa do
Brasil, com ampla distribuição e aspectos ecológicos que favorecem sua
utilização como espécie bioindicadora dos efeitos da poluição ambiental por
metais. Este trabalho teve por objetivo estudar MT e metais em acarás,
analisando a influência dos parâmetros biológicos do animal, além das
alterações produzidas pela exposição a um metal tóxico como cádmio (Cd).
Acarás coletados no rio Guandu foram divididos em dois grupos. O primeiro
considerado como não exposto e um segundo que foi exposto a Cd por injeção
intraperitoneal. MTs foram quantificadas por polarografia de pulso diferencial
enquanto zinco (Zn), cobre (Cu) e Cd em três frações hepáticas foram
quantificados por ICP OES. Os níveis de MT hepática e renal foram maiores em
acarás fêmeas e somente a MT branquial mostrou correlação positiva com peso
e tamanho corporal. O sexo não influenciou na concentração hepática de Zn e
Cu, entretanto os níveis de Zn estiveram positivamente relacionados ao peso e
tamanho corporal. O Cu não mostrou qualquer relação com estes parâmetros
biométricos. Os níveis de MT hepáticos estiveram relacionados com o conteúdo
de Zn e Cu neste órgão. Ambos os metais mostraram-se predominantemente
citosólicos, sendo que o Cu foi encontrado quase totalmente na fração que
contém a MT. Após exposição ao Cd, a MT renal e branquial apresentaram
níveis significativamente maiores, assim como se encontrou uma redução
significativa nos níveis de Zn e Cu hepático. A exposição ao Cd também
promoveu uma alteração na distribuição intracelular do Zn e do Cu onde um
menor percentual desses metais foi verificado na fração citosólica que contém a
MT. Neste estudo foi verificado que MT e metais em acarás são influenciados
por alguns parâmetros biológicos. A exposição ao Cd também promove uma
alteração nos níveis de MT e de metais bem como na distribuição intracelular.
Estes fatores estão relacionados com à toxicidade e a tolerância à exposição a
metais e devem ser considerados para que esta espécie possa ser utilizada como
bioindicadora da exposição ambiental a metais.
Abstract
Metallothioneins (MTs) are cytosolic metal-binding proteins whose biological
function are related with the regulation of essential metals, such Zn and Cu, and with
the detoxification of toxic metal such Cd and Hg. Knowledge on metal intracellular
compartmentalization, on the physiological function of MTs and on the factors that
control their levels is essential to understand the toxic effects induced by metals in fish
species. Geophagus brasiliensis (acará) is Brazilian native fish specie widely distributed
and with ecological and physiological characteristics that made it suitable to be used as
bioindicator fish specie to assess the effects of heavy metal pollution. The aim of this
work was to study MT and metals in G. brasiliensis analyzing the influence of
biological parameters and the alterations caused by the exposition to a toxic metal such
Cd. Animals were collected for Guandu River, in Rio de Janeiro State and were divided
into two groups. A first group, which was considered as non-exposed; and a second
group, which was exposed to Cd by intraperitonal injection. Hepatic MT, renal MT and
gill MT were quantified by differential pulse polarography while, Cu, Cd from three
hepatic fractions (total fraction, citosolic fraction and MT fraction) were quantified by
ICP OES. Levels of hepatic and renal MT were bigger in females and only MT from
gills showed a positive correlation with weight and body length. Sex did not affect the
hepatic levels of Zn and Cu, however levels of Zn were positively related with weigh
and body length. Cu did not show any relation with both these biological parameters.
Levels of hepatic MT were related with Zn and Cu contents of this organ. Both metals
showed to be mainly cytosolic, being Cu is largely found in the MT containing fraction.
After Cd exposition, renal and gill MT presented levels significantly higher, and it was
also found significant reduction in the levels of hepatic Zn and Cu. Exposition to Cd
caused also alterations on the pattern of intracellular distribution of Zn and Cu, since
less ratio of these metals were found in the MT containing fraction. Hence, this study
shows that MT and metals in G. brasiliensis are influenced by biological parameters and
that exposition to Cd causes alterations in the levels both of them and also alters the
patterns of metallic intracellular distribution. All these factors, that are related to
toxicity and tolerance to metal exposition, must be considered when this fish specie to
be used as bioindicator of environment exposition to heavy metals.
