Dada a complexidade do diagnóstico diferencial do vírus Zika (Zika virus - ZIKV) e a inexistência de propostas terapêuticas eficazes, nosso grupo de pesquisa previamente idealizou e engenheirou in silico três proteínas, denominadas VLL-V2, VLL-V3 e VLL-V4, a partir da estrutura do anticorpo monoclonal (monoclonal antibody – mAb) ZV-67, direcionado ao domínio DIII da proteína do envelope do ZIKV (E-ZIKV). As proteínas sintéticas foram desenvolvidas de modo a carrear os aminoácidos do mAb fundamentais na interação anticorpo-epítopo. Portanto, a hipótese deste trabalho é de que as proteínas desenvolvidas são capazes de se ligar ao seu alvo com alta afinidade e de neutralizar a infecção viral in vitro. Assim, o objetivo deste trabalho foi produzir in vitro as proteínas sintéticas e avaliá-las quanto à afinidade de ligação à proteína E-ZIKV e quanto à capacidade de neutralização da infecção viral. As proteínas foram expressas em células de E. coli, purificadas por cromatografia de afinidade, avaliadas quanto à interação com E-ZIKV através de termoforese em microescala (MST) e, por fim, o potencial das proteínas em neutralizar o ZIKV foi mensurado por ensaios de microneutralização viral em placa (MN). Além disso, o mAb 4G2 foi também produzido neste trabalho. Foram obtidas concentrações de 237,45 μM, 198,6 μM e 141,4 μM para as proteínas sintéticas VLL-V2, VLL-V3 e VLL-V4, respectivamente. Como resultado da MST, foi observado que a VLL-V2 apresentou a maior afinidade de ligação à E-ZIKV com uma constante de dissociação (KD) de 7,8 x 10-9 M. No ensaio de MN, foi demonstrado que a VLL-V2 foi capaz de neutralizar o ZIKV in vitro com uma EC50 de 0,04 μM. Logo, dentre as proteínas sintetizadas e testadas in vitro, a VLL-V2 apresentou uma maior afinidade de ligação à E-ZIKV, assemelhando-se aos valores apresentados por anticorpos neutralizantes já descritos na literatura. Adicionalmente, a proteína VLL-V2 apresentou potencial de neutralização da infecção por ZIKV in vitro .
Abstract
Given the complexity of the differential diagnosis of Zika virus (ZIKV) and the lack of effective therapeutic proposals, our research group engineered in silico three proteins (VLL-V2, VLL-V3 and VLL-V4) based on the structure of the ZV-67 monoclonal antibody (mAb) directed to DIII domain of ZIKV envelope protein (ZIKV-E). The synthetic proteins were developed in order to carry the essential mAb amino acids for the antibody-epitope interaction.Therefore, the hypothesis of this work is that the developed proteins are able to bind their target with high affinity and to neutralize the viral infection in vitro. The objective of this work was to produce in vitro the three synthetic proteins and evaluate them for their binding affinity to envelope protein of ZIKV, as well as the capacity to neutralize the viral infection. The proteins were expressed in E. coli cells, purified by affinity chromatography, evaluated for interaction with ZIKV-E by microscale thermophoresis (MST) and finally, the proteins ability in neutralize ZIKV infection was measured using plate viral microneutralization assays (MN). 4G2 mAbs were also produced in vitro in this work. After protein expression and purification, were obtained concentrations of 237,45 μM, 198,6 μM and 141,4 μM for synthetic proteins VLL-V2, VLL-V3 and VLL-V4, respectively. As a result of MST, it was observed that VLL-V2 had the highest binding affinity to ZIKV-E with a dissociation constant (KD) of 7.8 x 10-9 M. After microneutralization assays, it was demonstrated that the VLL-V2 protein was able to neutralize ZIKV in vitro with an EC50 of 0.04 μM. Among the proteins synthesized and tested in vitro, the VLL-V2 protein showed a high binding affinity to E protein, similar to the values presented by neutralizing antibodies already described in the literature. Additionally, the VLL-V2 protein also had potencial to neutralize ZIKV infection in vitro .
