Resumo | Os estudos voltados para entender a biologia da infecção causada pelo Zika vírus (ZIKV) em diferentes tecidos e espécies de mosquitos ainda são escassos. Para infectar uma célula hospedeira e iniciar a sua replicação, o vírus precisa ser internalizado no compartimento intracelular. O processo consiste em uma série intrincada de eventos altamente dinâmicos, fortemente coordenados, incluindo, entre outros, a ligação do vírus às células, o tráfico intracelular e a entrega da informação genética do vírus. A falta de conhecimentos sobre os mecanismos associados à biologia da infecção do mosquito vetor pelo ZIKV, incentivaram o presente estudo que investigou de forma comparativa a infecção do ZIKV em duas linhagens celulares de inseto com perfis distintos de suscetibilidade e permissibilidade à progressão do ciclo de infecção viral, a suscetível C6/36 (de Aedes albopictus) e a não suscetível Sf9 (de Spodoptera frugiperda). Desta forma, o objetivo central deste projeto foi caracterizar fatores moleculares e ultraestruturais determinantes para a internalização do vírus Zika PE243 em linhagens celulares de inseto, através da investigação da cinética de infecção viral, da avaliação ultraestrutural e do perfil transcriptômico de células C6/36 e Sf9 em resposta à infecção. A cinética de replicação viral foi avaliada por RT-qPCR e o monitoramento da internalização viral, bem como investigar possíveis alterações ultraestruturais induzidas pela infecção, foram avaliadas por microscopia eletrônica de varredura (MEV) e de transmissão (MET). As variações na expressão de genes envolvidos em vias metabólicas associadas à progressão do ciclo de infecção e patogênese viral foram investigadas através de RNAseq em equipamento Illumina MiSeq System. As análises da cinética de replicação nas células C636 e Sf9 evidenciaram que o ciclo de replicação viral após a infecção do ZIKV em Sf9 não progride, contrastando com a linhagem C6/36 cujo número de cópias de RNA viral progride significativamente em 48hpi (Cq de 24) e para Sf9 (Cq de 28) sem progressão de sinal de infecção independente do time point. Entretanto, através de microscopia MET e MEV foi demonstrado pela primeira vez alterações induzidas pela exposição do ZIKV na célula Sf9, como desorganização de membranas de retículo endoplasmático formando estruturas semelhantes a autofagossomos em ambas linhagens celulares, assim como a formação de corpos multivesiculares. Os resultados iniciais do sequenciamento forneceram informações gerais sobre a qualidade dos reads para as futuras análises. Os conhecimentos gerados contribuirão de forma significativa no entendimento da biologia da infecção do ZIKV em seus hospedeiros invertebrados. | en_US |

Resumo em Inglês | Studies aimed at understanding the biology of infection caused by the Zika virus (ZIKV) in different tissues and species of mosquitoes are still scarce. To infect a host cell and initiate its replication, the virus needs to be internalized into the intracellular compartment. The process consists of an intricate series of highly exciting, tightly coordinated events, including but not limited to virus attachment to cells, intracellular trafficking, and delivery of the virus's genetic information. The lack of knowledge about the controls associated with the biology of the mosquito vector infection by ZIKV encouraged the present study, which comparatively investigated the ZIKV infection in two insect cell lines with different profiles of susceptibility and permissibility to the progression of the infection cycle viral, the susceptible C6/36 (from Aedes albopictus) and the non-susceptible Sf9 (from Spodoptera frugiperda). Thus, the main objective of this project was to characterize molecular and ultrastructural factors that determine the internalization of the Zika virus PE243 in insect cell lines, through the investigation of viral infection kinetics, evaluation of the ultrastructural and transcriptomic profile of C6/36 and Sf9 in response to infection. The viral replication kinetics was evaluated by RT-qPCR and the monitoring of the viral internalization structure, as well as investigating possible ultranal changes induced by the infection, were evaluated by scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Variations in the expression of genes involved in metabolic pathways associated with the progression of the infection cycle and viral pathogenesis were investigated using RNAseq in the Illumina MiSeq System equipment. Analysis of the replication kinetics in C636 and Sf9 cells showed that the viral replication cycle after ZIKV infection in Sf9 does not progress, in contrast to the C6/36 lineage, whose number of viral RNA copies significantly progresses in 48hpi (Cq of 24) and for Sf9 (Cq of 28) with no infection signal progression regardless of time point. However, TEM and SEM microscopy demonstrated for the first time changes induced by exposure to ZIKV in the Sf9 cell, such as disorganization of endoplasmic reticulum membranes forming structures similar to autophagosomes in both cell lines, as well as the formation of multivesicular bodies. The initial sequencing results provide general information about the quality of the reads for future analyses. The knowledge generated will significantly contribute to the understanding of the biology of ZIKV infection in its invertebrate hosts. | en_US |
