Abstract in Portuguese | O potencial terapêutico de células da medula óssea tem sido bastante investigado no campo da medicina regenerativa. Embora os resultados iniciais de ensaios pré-clínicos e clínicos com células da medula óssea em doenças hepáticas sejam bastante promissores, os efeitos terapêuticos não são duradouros. Assim, maiores estudos vêm sendo realizados com o objetivo de otimizar a terapia celular. Dentre as populações de células-tronco, as células-tronco mesenquimais (CTMs) se destacam por apresentarem plasticidade genômica e características imunomoduladoras e imunossupressoras que ampliam as possibilidades de utilização terapêutica. Um dos fatores imunomodulatórios secretados pelas CTMs é o fator de crescimento de hepatócitos (HGF), cuja participação em processos antifibróticos e antiinflamatórios já é descrita. Diante disso, o presente trabalho objetivou avaliar o potencial antifibrogênico de CTMs superexpressando HGF na fibrogênese hepática. Para atingir os objetivos propostos, as CTMs foram obtidas de medula óssea de camundongos C57BL/6 e caracterizadas.O RNA total foi extraído de tecido hepático de camundongos da mesma linhagem para obtenção do gene Hgf que foi amplificado por PCR, clonado no vetor de expressão pCMV-LacZ e transfectado nas CTMs. A transfecção se mostrou eficiente pela análise da expressão do Hgf feita por RT-PCR. As CTMs modificadas foram co-cultivadas por 48h com células estreladas hepáticas (CEHs) da linhagem GRX. citocinas e a expressão do marcador da fibrogênese hepática, alfa-actina de músculo liso (α-SMA) foram quantificados, onde foi observado um aumento na secreção de IL-6 e redução de TGFβ1. Os baixos níveis de α-SMA observados, juntamente com os achados da caracterização ultraestrutural e molecular da linhagem GRX, demonstraram que estas células podem não se encontrar no seu fenótipo ativado, reforçando a necessidade de uma observação mais criteriosa para a utilização desta linhagem em estudos in vitro da fibrogênese hepática. | pt_BR |

Abstract | The therapeutic potential of bone marrow cells has been extensively investigated in the field of regenerative medicine. Regarding liver disease, although initial results from preclinical and clinical trials using bone marrow cells are quite promising, the therapeutic effects are not long-lasting. Thus, more studies have been carried out with the aim of optimizing cell therapy. Among the populations of stem cells, the mesenchymal stem cells (MSCs) stand out because they present genomic plasticity and immunomodulatory and immunosuppressive characteristics that increase the possibilities of therapeutic use. One of the immunomodulatory factors released by MSCs is the hepatocyte growth factor (HGF), whose participation in antifibrotic and anti-inflammatory processes is already described. Thus, the present work aimed to evaluate the antifibrogenic potential of MSCs overexpressing HGF in hepatic fibrogenesis. To achieve the proposed objectives, the MSCs were obtained from bone marrow of C57BL/6 mice and characterized.Total RNA was extracted from livers from the same lineage mice to obtain the Hgf gene that was amplified by PCR, cloned in the expression vector pCMV-LacZ and transfected into the MSCs.The transfection proved to be efficient by the analysis of HGF expression by RT-PCR. Modified CTMs were co-cultured for 48h with hepatic stellate cells (CEHs) of the GRX lineage and cytokine and hepatic fibrogenesis marker expression, smooth muscle actin – α (α-SMA), were evaluated and was observed an increase in IL-6 secretion and reduction of TGFβ1. The low levels of α-SMA observed, together with the findings of the ultrastructural and molecular characterization of the GRX lineage, demonstrated that these cells may not be in their activated phenotype, reinforcing the need of a careful observation for the use of this lineage in vitro studies of hepatic fibrogenesis. | pt_BR |
