Abstract in Portuguese | Análises de genômica comparativa mostraram que a conservação dos componentes desta via de exportação de mRNA em espécies de parasitas é baixa quando comparada a outras vias de exportação nuclear. Uma RNA Helicase tipo DEAD box, denominada UAP56 (mamíferos)/Sub2(leveduras), é a mais conservada ao longo da filogenia de eucariotos, sendo essencial para a exportação de mRNA. Ensaios de RNAi em T.brucei demonstraram que essa proteína (TbSub2) também é essencial para a exportação de mRNA em tripanosomatídeos. Contudo, apesar de ser bem conservada, não podemos concluir que esta proteína tenha a mesma função em outras espécies de tripanosomatídeos. Infelizmente, não foi possível obter linhagens para o nocaute do gene (TcSub2) em T.cruzi, nosso modelo de estudo, provavelmente por ser tratar de uma proteína essencial à sobrevivência do parasita. Por isso, o objetivo deste trabalho foi estudar a função e estrutura da proteína TcSub2 através de ensaios enzimáticos, espectroscopia de dicroísmo circular, cromatografia de exclusão molecular, modelagem molecular e complementação funcional em T.brucei. Esta proteína pertence à família de helicases DEAD box que manipulam RNAs estruturados em complexos de proteína-RNA utilizando hidrólise de ATP como fonte de energia. Os ensaios bioquímicos demonstraram que o domínio ATPase de TcSub2 é funcional e a atividade enzimática
influenciada pela ligação ao RNA, independente de sequência do RNA. A caracterização funcional e estrutural da proteína mutante, TcSub2K87N, provou que o resíduo conservado K87 é essencial para a hidrólise de ATP e que sua substituição por uma Asparagina não altera sua estrutura secundária e quaternária. A ausência de atividade é devido, provavelmente, à pequenas alterações na estrutura tridimensional que impossibilitam o correto posicionamento do ATP no sítio ativo de TcSub2. Para os ensaios de complementação funcional, foi estabelecida uma linhagem de T. brucei capaz de silenciar a expressão de TbSub2 por RNAi. Esta linhagem será então utilizada para ensaios futuros visando avaliar a semelhança de

Abstract | Comparative genomic analyzes have showed that components mRNA export pathway are low conserved in species of parasite as compared to components from other nuclear export pathways. However, the RNA Helicase DEAD box, named UAP56 (mammals) / Sub2 (yeast), is the highest conserved through phylogeny of eukaryotes and essential for the
mRNA export. RNAi assays in T. brucei have shown that this protein (TbSub2) is also essential for the mRNA export in trypanosomes. However, despite of highly conserved, we cannot conclude that this protein has the same function in other species of trypanosomes. Unfortunately, it was not possible to obtain a knockout line for TcSub2 gene in T. cruzi, our study model, since it might be essential for survival of the parasite. Therefore, the main goal of this work was to study the function and structure of TcSub2 protein by enzymatic assays, circular dichroism spectroscopy, molecular exclusion chromatography, molecular modeling and functional complementation in T. brucei. This protein is member of DEAD box helicases family that handling structured RNAs in RNA-protein complexes using ATP hydrolysis as an energy source. Biochemical assays have demonstrated that the TcSub2 ATPase domain is functional and RNA binding but not RNA sequence has modified the enzymatic activity. The
structural and functional characterization of mutant protein, TcSub2K87N, have proved that the conserved residue K87 is essential for ATP hydrolysis and the exchange for asparagine does not alter its secondary and quaternary structure. The lack of activity is probably due to small changes in tridimensional structure that preclude the correct binding of ATP in the
active site of TcSub2. For functional complementation assays, a RNAi T. brucei line has been established to knockdown the TcSub2 expression. It will be useful for further analysis in order to evaluate the similarity of function and activity between T. brucei and T. cruzi proteins. | pt_BR |
