As leishmanioses estão entre as doenças mais negligenciadas do mundo. São causadas por
protozoários do gênero Leishmania cujos vetores são flebotomíneos (Ordem Diptera; Família
Psychodidae; subfamília Phlebotominae). Estudos realizados em áreas endêmicas de
leishmaniose tegumentar e visceral tem demonstrado a presença do flebotomíneo Lutzomyia
migonei como importante vetor na transmissão da leishmaniose tegumentar e, na ausência de
Lutzomyia longipalpis, também da leishmaniose visceral. Apesar de a literatura apresentar
muitos estudos sobre a fauna flebotomínica, muitos aspectos relacionados a resposta imune
dos flebótomos vetores ainda permanecem desconhecidos. Sendo assim, o presente estudo tem
por objetivou quantificar e caracterizar os hemócitos envolvidos na resposta imune celular
inata de Lu. migonei, assim como fazer o levantamento da fauna flebomínica no município de
Timbaúba, área endêmica de leishmaniose tegumentar de Pernambuco. Os insetos foram
mantidos no Laboratório de Imunopatologia Keizo Asami-LIKA-UFPE/PE. A coleta da
hemolinfa para caracterização dos hemócitos foi realizada com um micromanipulador. As
células foram quantificadas e caracterizadas através da microscopia de campo claro e da
microscopia eletrônica de transmissão e foram classificadas como prohemócitos, granulócitos,
adipohemócitos, plasmatócitos e eonocitóides. Já a identificação dos espécimes utilizou-se de
chave taxonômica e classificou os flebotomíneos em seis espécies: Lutzomyia whitmani, Lu.
migonei, Lu. evandroi, Lu. wellcomei, Lu. quinquefer e Lu. sordellii.
Abstract
Leishmaniasis is among the most neglected diseases in the world. They are caused by
protozoa of the genus Leishmania whose vectors are phlebotomines (Diptera Order, Family
Psychodidae, subfamily Phlebotominae). Studies in endemic areas of tegumentary and
visceral leishmaniasis have demonstrated the presence of phlebotomine Lutzomyia migonei as
an important vector in the transmission of tegumentary leishmaniasis and, in the absence of
Lutzomyia longipalpis, also of visceral leishmaniasis. Although the literature presents many
studies on phlebotomine fauna, many aspects related to the immune response of the
phlebotomine vectors still remain unknown. Thus, the present study aims to quantify and
characterize the hemocytes involved in the innate cellular immune response of Lu. migonei, as
well as the survey of the phlebominal fauna in the municipality of Timbaúba, an endemic area
of tegumentary leishmaniasis of Pernambuco. The insects were kept in the Laboratory of
Immunopathology Keizo Asami-LIKA-UFPE / PE. Hemolymph collection for haemocyte
characterization was performed with a micromanipulator. Cells were quantified and
characterized by light field microscopy and transmission electron microscopy and were
classified as prohemocytes, granulocytes, adipohemocytes, plasmatocytes and eonocites. The
identification of the specimens was used as a taxonomic key and classified the sandflies into
six species: Lutzomyia whitmani, Lu. migonei, Lu. evandroi, Lu. wellcomei, Lu. quinquefer
and Lu. sordellii.
