Com base na idéia de que esporos bacterianos são estruturas extremamente resistentes, fomos capazes a partir da utilização de várias técnicas de cultivar e identificar de coprólitos humanos e animais, datados entre 3490 ± 120 a 430 ± 70 anos, recuperados de escavações arqueológicas no Brasil, bactérias da família Bacillaceae. Os coprólitos inicialmente foram tratados com radiação ultravioleta com o objetivo de eliminar possíveis contaminantes externos e posteriormente perfurados assepticamente para retirada de material da porção mais interna. Esse material foi então cultivado emdiferentes meios de cultura, possibilitando o isolamento de bastonetes Gram positivos. Esses microorganismos foram então submetidos a reações bioquímicas clássicas e a técnicas moleculares, como a reação em cadeia da polimerase (PCR), a clonagem e o seqüenciamento gênico. A análise de todos os dados possibilitou não somente concluir que se tratavam de representantes da família Bacillaceae, mas em alguns casos definir ogênero e a espécie mais provável, demonstrando que estas metodologias conjugadas permitem este grau de definição e que será possível no futuro um estudo mais minucioso da microbiota antiga associada a este material.
