Abstract in Portuguese | As leishmanioses são doenças parasitárias de origem zoonótica transmitidas por insetos flebotomíneos. A distribuição destas doenças está diretamente relacionada à presença, expansão e dispersão dos flebotomíneos. O presente estudo teve como objetivo estudar a fauna de flebotomíneos na área urbana de Lassance, detectar DNA de Leishmania nas fêmeas coletas, aferir sua capacidade de voo e estudar os aspectos taxonômicos relacionados à espécie Lutzomyia longipalpis, principal espécie envolvida na veiculação da Leishmania infantum no Brasil. Foram realizadas coletas com armadilhas luminosas do tipo HP na área urbana do município de Lassance - MG entre os meses de Maio de 2018 e Abril de 2019. Nas fêmeas coletadas foi realizada análise molecular para determinar a presença de DNA de Leishmania através da técnica de PCR, dirigida ao alvo ITS1. A identificação da espécie de Leishmania envolvida na detecção foi realizada através da técnica de PCR-RFLP HaeIII. Para estudo da capacidade de voo foram instaladas armadilhas na região central do rio das Velhas, a 80 metros equidistantes entre si e entre cada margem. Para o estudo taxonômico, flebotomíneos machos do complexo Lu. longipalpis, de 4 localidades (Campo Grande, Jacobina, Sobral e Lassance) foram utilizados para análise de ultramicroscópica de estruturas cuticulares internas que podem estar associadas à produção de feromônio. Um total de 1.611 espécimes de flebotomíneos foi capturado na área urbana de Lassance, com predominância das espécies Lu. longipalpis e Nyssomyia intermedia. DNA de Le. infantum em Lu. longipalpis foi detectado na área urbana de Lassance. No estudo da capacidade de voo, foi capturado um total de 804 espécimes de flebotomíneos sendo 798 indivíduos nas margens e 6 indivíduos na região central do rio, comprovando que os flebotomíneos podem voar de forma ininterrupta por pelo menos 80 metros. As análises de microscopia de varredura revelaram estruturas internas responsáveis pela liberação de feromônio no complexo Lu. longipalpis, sendo denominadas "manifold". Além disso, foi possível observar diferenças morfológicas nesta estrutura, entre as populações deste complexo de espécies que produzem diferentes quimiotipos. Os resultados encontrados aqui auxiliarão nas medidas de controle vetorial e compreensão da epidemiologia das leishmanioses no município de Lassance além de fornecer dados sobre a capacidade de voo dos flebotomíneos e melhor entendimento sobre a taxonomia do complexo Lu. longipalpis no que tange as estruturas relacionadas a liberação de feromônio. | pt_BR |

Abstract | Leishmaniases are parasitic diseases of zoonotic origin transmitted by sand flies. The distribution of these diseases is directly related to the presence, expansion and dispersion of sand flies. This study aimed to study the sand fly fauna in the urban area of Lassance, detect Leishmania DNA in collected females, assess their flight capacity and study the taxonomic aspects related to the species Lutzomyia longipalpis, the main species involved in the dissemination of Leishmania infantum in the Brazil. Collections were carried out with HP type light traps in the urban area of the municipality of Lassance - MG between the months of May 2018 and April 2019. In the collected females a molecular analysis was performed to determine the presence of Leishmania DNA through the PCR technique, directed at the ITS1 target. The identification of the Leishmania species involved in the detection was performed using the PCR-RFLP HaeIII technique. To study the flight capacity, traps were installed in the central region of the Velhas’ river, 80 meters equidistant from each other and between each bank. For the taxonomic study, male sand flies from the Lu. longipalpis complex, from 4 locations (Campo Grande, Jacobina, Sobral and Lassance) were used for ultramicroscopic analysis of internal cuticular structures that may be associated with pheromone production. A total of 1,611 sandfly specimens were captured in the urban area of Lassance, with a predominance of the Lu. longipalpis species and Nyssomyia intermedia. DNA of Le. infantum in Lu. longipalpis was detected in the urban area of Lassance. In the study of flight capacity, a total of 804 specimens of sand flies were captured, 798 individuals on the banks and 6 individuals in the central region of the river, proving that sand flies can fly uninterruptedly for at least 80 meters. Scanning microscopy analyzes revealed internal structures responsible for pheromone release in the Lu. longipalpis complex, being called "manifold". Furthermore, it was possible to observe morphological differences in this structure among populations of this complex of species that produce different chemotypes. The results found here will help to measure vector control and understand the epidemiology of leishmaniasis in the municipality of Lassance, in addition to providing data on the flight capacity of sand flies and a better understanding of the taxonomy of the Lu. longipalpis complex regarding structures related to pheromone release. | pt_BR |
