Corpúsculos lipídicos (CLs) são organelas citoplasmáticas responsáveis pelo armazenamento de lipídios neutros e pelo metabolismo de mediadores em muitos tipos de células. Dados recentes mostram formação de CLs e produção de eicosanoides em Leishmania. Contudo, estudos da formação e modulação de CLs, bem como, da síntese de lipídios bioativos em diferentes espécies de Leishmania ainda não foram conduzidos. Assim, primeiramente foram avaliadas por microscopia ótica a formação de CLs em L. infantum, L. amazonensis e L. braziliensis, bem como, a modulação destas organelas estimuladas com os ácidos eicosapentaenóico (EPA), docosahexaenoico (DHA) e araquidônico (AA). Os mediadores produzidos por estas espécies foram quantificados por cromatografia líquida acoplada à espectrometria de massas (HPLC-MS) e a expressão das proteínas GP63 e PGFS foram avaliadas por imunoblotting e ELISA. Por fim, as sequencias genômicas e os resíduos do sítio ativo dessas proteínas foram analisadas em espécies de Leishmania do Velho e Novo Mundo. Observamos maior número de CLs durante metaciclogênese e após estimulação com EPA, DHA e AA, em L. infantum e L. braziliensis. Além disso, identificamos 12 lipídios bioativos, estimulados principalmente por AA. A partir das análises in silico, notamos que as sequências de GP63 e PGFS são mais similares entre as espécies de mesmo tropismo e que os resíduos do sítio ativo de GP63 foram mais alterados, ao contrário dos resíduos de PGFS. Também observamos que GP63 foi mais produzida na fase logarítmica dos parasitos, ao contrário de PGFS que foi mais expressa na fase estacionária. No contexto da interação parasito-hospedeiro, ainda não foi investigado a síntese de eicosanoides in situ durante a fase crônica da leishmaniose visceral (LV). Desse modo, utilizamos Hamsters Golden Syrian (Mesocricetus auratus) machos, com 6 a 8 semanas de idade, que foram infectados intraperitonealmente com 107 L. infantum/mL. Após 150 dias de infecção, avaliamos as alterações clínicas e histológicas no baço e fígado. Através da HPLC-MS/MS demonstramos pela primeira vez a produção de eicosanoides no plasma, baço e fígado dos animais. Notamos que o aumento da produção dos mediadores HETEs está correlacionado com a gravidade da LV. Portanto, este estudo abre perspectivas para compreensão da biologia de Leishmania e sua interação com o hospedeiro, bem como amplia o conhecimento para elaboração de novas estratégias antiparasitárias

Lipid droplets (LDs) are cytoplasmic organelles responsible for the storage of neutral lipids and the mediator’s metabolism in many types of cells. Recent data show LDs formation and eicosanoid production in Leishmania. However, studies on the formation and modulation of LDs, as well as the bioactive lipids synthesis in different Leishmania species have not yet been conducted. Thus, the formation of LDs in L. infantum, L. amazonensis and L. braziliensis, as well as the stimulation of these organelles with eicosapentaenoic (EPA), docosahexaenoic (DHA) and arachidonic (AA) acids were first evaluated by optical microscopy. The mediators produced by these species were quantified by High-Performance Liquid Chromatography - Mass Spectrometry (HPLC-MS) and the expression of GP63 and PGFS proteins were evaluated by immunoblotting and ELISA. Finally, the genomic sequences and active site residues of these proteins were analyzed in Old and New World Leishmania species. We observed a greater number of LDs during metacyclogenesis and after stimulation with EPA, DHA and AA, in L. infantum and L. braziliensis. In addition, we identified 12 bioactive lipids, mainly stimulated by AA. From the in-silico analyses, we noticed that GP63 and PGFS sequences are more similar between species with the same tropism and that GP63 active site residues were more altered, unlike the PGFS residues. We also observed that GP63 was more produced in the log phase of the parasites, unlike PGFS which was more expressed in the stationary phase. In the context of the parasite-host interaction, the eicosanoids synthesis in situ during the chronic phase of visceral leishmaniasis (VL) has not yet been investigated. Therefore, we used male Golden Syrian Hamsters (Mesocricetus auratus), aged 6 to 8 weeks, which were infected intraperitoneally with 107 L. infantum/mL. After 150 days of infection, we evaluated the clinical and histological changes in the spleen and liver. Through HPLC-MS, we demonstrated for the first time the eicosanoids production in the plasma, spleen and liver of animals. We noticed that the increase in the production of HETEs mediators is correlated with the severity of VL. Therefore, this study opens perspectives for understanding the biology of Leishmania and its interaction with the host, as well as expanding the knowledge for the development of new antiparasitic strategies
