Abstract in Portuguese | A cólera é uma doença reemergente que tem como agente etiológico o Vibrio cholerae O1,
biotipo El Tor. Atualmente a doença encontra-se sob controle no país, no entanto, o risco de
sua re-introdução continua presente tendo em vista que persistem as baixas coberturas de
saneamento e o V. cholerae O1 continua sendo isolado de águas ambientais, mesmo
esporadicamente. Apesar de já terem sido identificados mais de 200 sorogrupos de V. cholerae,
somente os sorogrupos O1 e O139 são responsáveis por epidemias. Os demais sorogrupos
podem ser patogênicos e estar associados a surtos de diarréia. A patogenicidade de V. cholerae
epidêmico está relacionada a duas regiões do cromossomo maior do vibrio, o elemento
genético CTX e a ilha de patogenicidade de Vibrio (VPI) que devido as suas características de
bacteriófago podem ser transferidos horizontalmente conferindo virulência às cepas receptoras.
Neste estudo descrevemos a caracterização parcial dos elementos genéticos CTX e VPI de
cepas de V. cholerae O26 isoladas no Brasil entre os anos de 1992 e 2000. Através da
amplificação dos genes cep, orfU, ace, zot, cxtA, ctxB, rstA, rstB, rstR presentes no profago
CTXφ foi visto que oito cepas possuem o profago CTXφ completo e sete o profago incompleto
(pré-CTX). Os genes aldA, tagA, tcpPH e int das regiões TAG, ACF e TCP da VPI estão
presentes na maioria das cepas, no entanto, somente duas (151 e 168) apresentam o gene tcpA
El Tor. O sequenciamento dos genes rstR do profago CTXφ e tcpA da VPI mostrou que o tipo
e alelo para os dois genes detectados na maioria das cepas O26 corresponde a variante presente
em cepas ambientais de sorogrupos não-O1/ não-O139. Os alelos presentes nas cepas 151 e
168 correspondem ao do V. cholerae O1 El Tor. Conclui-se que há especificidade entre o tipo
de gene tcpA e o profago CTXφ de cada cepa. Nesse estudo identificamos a existência de cepas
de V. cholerae não-O1/ não-O139 isoladas no Brasil que possuem todo o conjunto de genes de
virulência presentes nos sorogrupos epidêmicos de V. cholerae e que são negligenciadas no
diagnóstico tradicional da cólera. Desta forma, recomenda-se que além dos testes de triagem
tradicionais, indispensáveis para a classificação do V. cholerae, sejam realizados métodos
moleculares complementares que permitam a identificação de cepas carreando genes de

Abstract | Cholera is a reemerging disease that has as the etiological agent the Vibrio cholerae O1,
biotype E1 Tor. Currently, the disease has been kept under control in our country, however,
the risk of its re-introduction remains, since the low sanitation services are common and the V.
cholerae O1 keeps on being isolated from environmental waters, although occasionally. Even
though more than 200 serogroups have already been identified, only the serogroups O1 and
O139 are responsible for the epidemics. The other serogroups can be pathogenic and be
related to diarrhea outbreaks. The pathogenicity of the epidemic V. cholerae is related to two
regions in the large chromossome of the vibrio, the genetic element CTX and the Vibrio
pathogenicity island (VPI), that due to its bacteriophage characteristics can be transferred
horizontally conferring virulence to the receptor strains. In this study, we describe the partial
characterization of the genetic elements CTX and VPI from V. cholerae O26 strains isolated
in Brazil, from 1992 to 2000. Through the amplification of the genes cep, orfU, ace, zot, cxtA,
ctxB, rstA, rstB, rstR present in the CTXφ it could be seen that eight strains have the CTXφ
complete prophage and seven the incomplete prophage (pre-CTX). The genes aldA, tagA,
tcpPH e int from the regions TAG, ACF e TCP from VPI, are present in most strain, however,
only two strains (151 and 168) present the tcpA gene E1 Tor. The sequencing of the rstR
genes of the CTXφ prophage and the tcpA of the VPI showed that the type and allele for the
two genes detected in most O26 strains corresponds to the variant present in environmental
strains of non-O1/non-O139 serogroups. The alleles present in strains 151 and 168 correspond
to the O1 E1 Tor. It is concluded that there is specificity between the type of tcpA and
prophage CTXφ of each strain. Therefore, we identify in this study the existence of non-
O1/non-O139 V. cholerae strains isolated in Brazil, holder of the whole set of virulence genes
present in epidemic V. cholerae serogroups. Thus, it is recommended that screening tests
further on traditional tests, essential for the classification of V. cholerae are carried out
complementary molecular methods to identify the strains carried virulence genes. | en_US |
