Abstract in Portuguese | Introdução: Este estudo teve por objetivo realizar um amplo e premente levantamento dos riscos da matéria em escala nanométrica (10-9 ), a luz da Nanotoxicologia, com ênfase no ta manho de 1 a 100 nm, em especial das nanopartículas de prata (AgNPs), empreendendo uma avaliação da qualidade de Revisões Sistemáticas (RS) sobre a toxicidade destes materiais a curto e longo prazo. O aumento recente da produção de nanomateriais (NMs) exige uma mai- or investigação sobre sua toxicidade potencial e efeitos adversos para a saúde após a exposi- ção. A eleição da AgNP, dentre outras, se deve à relevância de tais potenciais efeitos adversos gerados, seu volume de produção mundial e ao contingente de literatura científica publiciza da. Objetivos: Avaliar a qualidade de RS sobre a toxicidade das AgNPs em células in vitro, in vivo e ex vivo, em mamíferos, incluindo humanos, abrangendo metodologias e vieses po tencialmente presentes, discutindo potenciais repercussões e necessidades de controle. Méto do: A metodologia escolhida foi a avaliação da qualidade (Quality Assessment) de RS que versam sobre a toxicidade dos NMs, sobretudo a nanoprata. Para tal, foi conduzida uma ampla pesquisa eletrônica sem restrições de data ou idioma, em vários bancos de dados. Os critérios de elegibilidade incluíram RS, com ou sem metanálise, focadas na toxicidade de nanopartícu las de prata. Duas importantes ferramentas foram utilizadas para avaliar a qualidade destes estudos, quais sejam, AMSTAR 2, que avalia a qualidade metodológica de tais revisões, e ROBIS, para avaliação do risco de viés. Após o processo de seleção, 7 revisões sistemáticas foram incluídas. Cabe ressaltar que não foram encontrados Quality Assessments relacionados ao tema. Resultados: Dentre as sete revisões incluídas no estudo, especificamente, no que concerne à questão da pesquisa, pudemos observar diversidades no tema, evidenciando hete rogeneidade. Cinco revisões incluíram estudos experimentais e duas abrangeram estudos ob servacionais em humanos. Nenhuma das avaliações incluídas contemplou todos os critérios da ferramenta AMSTAR 2, evidenciando predomínio de grau de confiança criticamente baixo. Através da ferramenta ROBIS, constatamos três revisões avaliadas como “baixo potencial de viés”, três revisões consideradas de “alto potencial de viés” e duas de classificação “incerta”. Em geral, houve consistência entre ROBIS e AMSTAR. Conclusões: No presente Quality Assessment, nenhuma das revisões alcançou todos os critérios nos checklists utilizados. Reve la-se, portanto, uma irrefutável indicação da premência de padronização metodológica em futuras revisões. | en_US |

Abstract | Introduction: The aim of this study was to carry out an extensive and urgent study of nano metric scale (10-9 ), at the light of Nanotoxicology, with an emphasis on the size of 1 to 100 nm, especially silver nanoparticles (AgNPs), undertaking an evaluation of the quality of Sys tematic Reviews (SR) on the toxicity of these materials in the short and long term. The recent increase in the production of nanomaterials (NMs) requires further investigation into their potential toxicity and adverse health effects after exposure. The election of AgNP, among oth ers, is due to the relevance of such potential adverse effects generated, its volume of world production and the contingent of published scientific literature. Objectives: To assess RS quality on the toxicity of AgNPs in cells in vitro, in vivo and ex vivo, in mammals, including humans, covering potentially present methodologies and biases, discussing potential repercus sions and control needs. Method: The methodology chosen was the Quality Assessment of RS, which deals with the toxicity of NMs, especially nanosilver. For this, an extensive elec- tronic search was carried out without date or language restrictions, in the most diverse data- bases. Eligibility criteria included RS, with or without meta-analysis, focused on the toxicity of silver nanoparticles. Two important tools were used to evaluate the quality of these studies, namely AMSTAR 2, which evaluates the methodological quality of such reviews and ROBIS for bias risk assessment. After the selection process, 7 systematic reviews were included. It should be noted that Quality Assessments related to the theme were not found. Results: Among the seven reviews included in the study, specifically, regarding the research question, we could observe diversity in the theme, showing heterogeneity. Five reviews included exper- imental studies and two covered observational studies in humans. None of the included evalu- ations met all criteria of the AMSTAR 2 tool, evidencing a predominance of critically low degree of confidence. Through the ROBIS tool, we verified three reviews evaluated as "low bias potential", three reviews considered "high bias potential" and two "uncertain" rating. In general, there was consistency between ROBIS and AMSTAR. Conclusions: In the present Quality Assessment, none of the revisions met all the criteria in the checklists used. Therefore, there is an irrefutable indication of the urgency of methodological standardization in future reviews. | en_US |
