Abstract in Portuguese | O vírus Zika (Zika virus, ZIKV) é um arbovírus membro da família Flaviviridae e gênero
Flavivirus. Seu genoma é de RNA de sentido positivo e constituído por única matriz de
leitura, responsável pela tradução das proteínas virais estruturais e não estruturais, ladeada por
terminações não codificantes nas extremidades 5ˈ e 3ˈ. Atualmente, a ausência de vacinas ou
fármacos anti-ZIKV licenciados e o número de casos reportados de infecções por ZIKV, além
das complicações neurológicas decorrentes das infecções pelo vírus, colocam as infecções por
ZIKV como um grande problema de saúde pública, sobretudo em regiões tropicais e
subtropicais. Diante disso, o presente trabalho objetivou a construção de uma linhagem
celular expressando estavelmente um subgenoma autoreplicativo (replicon) do ZIKV,
expressando o gene repórter luciferase firefly (FLuc). Para tal, foi utilizado o genoma do
ZIKV cepa ZIKVPE243/2015, previamente isolado em Pernambuco e clonado no vetor
pSVJS01. A construção foi dividida estrategicamente em 3 fragmentos: i) o primeiro,
abrigando a extremidade 5ˈUTR e a sequência do capsídeo do ZIKV; ii) o segundo, formado
pelo gene repórter FLuc, uma região de ubiquitinação, gene da neomicina fosfotransferase e o
IRES (Internal ribosome entry site) do vírus da encefalomiocardite; iii) o último fragmento,
formado pelas proteínas não estruturais do ZIKV. Após a recombinação homóloga, a
identidade do replicon, denominado de repZIKVPE243-LucNeoIres, foi confirmada através
de PCR e sequenciamento. Transcritos in vitro produzidos a partir do cDNA clonado foram
transfectados em células BHK-21 por eletroporação. Três dias após a transfecção, as células
foram selecionadas em meio com geneticina (600µg/mL) e a linhagem celular recombinante
obtida, BHK-21-rZIKV-LucNeoIres, foi avaliada quanto à expressão de FLuc. Durante 20
passagens, as células BHK-21-rZIKV-LucNeoIres apresentaram um aumento da atividade da
FLuc de no mínimo 20 vezes em relação ao controle negativo. Por fim, a linhagem celular
recombinante BHK-21-rZIKV-LucNeoIres poderá ser usada para o desenvolvimento de uma
plataforma biotecnológica para triagem em larga escala de compostos anti-ZIKV e em estudos

Abstract | The Zika virus (Zika virus, ZIKV) is an arbovirus member of the family Flaviviridae and
genus Flavivirus. Its genome is positive-sense RNA and consists of a single reading matrix,
responsible for the translation of structural and non-structural viral proteins, flanked by
noncoding endings at ends 5 and 3. Currently, the absence of licensed anti-ZIKV vaccines or
drugs and the number of reported cases of ZIKV infections, in addition to the neurological
complications resulting from the virus infections, place ZIKV infections as a major public
health problem, especially in tropical regions and subtropical regions. Therefore, the present
work aimed to construct a cell line stably expressing a ZIKV autoreplicative subgenome
(replicon), expressing the reporter gene luciferase firefly (FLuc). For this, the genome of the
ZIKV strain ZIKVPE243 / 2015, previously isolated in Pernambuco and cloned in vector
pSVJS01, was used. The construct was strategically divided into 3 fragments: i) the first,
harboring the 5’UTR end and the ZIKV capsid sequence; ii) the second, formed by the FLuc
reporter gene, a ubiquitination region, neomycin phosphotransferase gene and the IRES
(Internal ribosome entry site) of encephalomyocarditis virus; iii) the last fragment, formed by
the non-structural proteins of ZIKV. After homologous recombination, the identity of
replicon repZIKVPE243-LucNeoIres was confirmed by PCR and sequencing. In vitro
transcripts produced from the cloned cDNA were transfected into BHK-21 cells by
electroporation. Three days after transfection, the cells were selected in medium with
geneticin (600μg / ml) and the recombinant cell line obtained, BHK-21-rZIKV-LucNeoIres,
was evaluated for FLuc expression. During 20 passages, BHK-21-rZIKV-LucNeoIres cells
showed an increase in FLuc activity of at least 20-fold compared to the negative control.
Finally, the recombinant BHK-21-rZIKV-LucNeoIres cell line could be used in the
