Os vírus Zika (ZIKV) e o vírus Chikungunya (CHIKV) podem causar diversas manifestações neurológicas graves. O ZIKV é capaz de gerar meningite e encefalite e em mulheres grávidas, pode levar ao desenvolvimento de microcefalia, malformações congênitas e morte fetal. O CHIKV apesar de não ser classificado como um vírus preferencialmente neurotrópico causa encefalite, meningite e a síndrome de Guillain-Barré. Recentemente, o cultivo de células em sistemas 3D foi capaz de gerar estruturas que mimetizam in vitro um microambiente de organização celular que ocorre in vivo. Nesse contexto, as células humanas de neuroblastoma, SH-SY5Y, representam um modelo celular promissor para o cultivo em 3D. Não está totalmente esclarecido como o ZIKV e CHIKV atuam na neurogênese, na qual estão envolvidos genes reguladores do desenvolvimento, como os genes HOX (Homeobox), que participam na diferenciação de diversos tipos de células. Com isso, o objetivo geral do estudo foi desenvolver novos modelos de cultivo neuronal em 3D (neuroesferas) a partir de células SH-SY5Y e avaliar a modulação da expressão dos genes HOX nestes modelos em processo de diferenciação celular após a infecção com ZIKV e CHIKV. Para isso, foi realizado o cultivo tridimensional de células SH-SY5Y, utilizando a técnica de levitação magnética. As neuroesferas foram diferenciadas em neurônios maduros com ácido retinóico all trans e BDNF (Brain derived neurotrophic factor) e infectadas com ZIKV e/ou CHIKV. Foi analisada por RT-qPCR a expressão dos genes HOX e por citometria de fluxo o perfil inflamatório das neuroesferas infectadas. Foi observado que as neuroesferas são capazes de se diferenciar em neurônios, são permissivas à infecção por ambos os vírus e geram resposta inflamatória com aumento dos níveis de IL-6 e IL-8 tanto para ZIKV quanto para CHIKV. A infecção por estes vírus altera de forma significativa a expressão dos genes SIX2, SIX3, SIX4 e SIX6 envolvidos com o processo de diferenciação celular. Outros genes HOX, como ARX, HOXD1, DMBX1, POU5F1 tiveram sua expressão significativamente alterada a depender do vírus. Estes resultados sugerem que neuroesferas são modelos viáveis para o estudo da neuropatogênese viral e a alteração da expressão dos genes HOX revela aspectos importantes desses vírus no percurso da neurogênese.
Abstract
Zika virus (ZIKV) and Chikungunya virus (CHIKV) can cause a variety of severe neurological manifestations. ZIKV is capable of generating meningitis and encephalitis and in pregnant women, it can lead to the development of microcephaly, congenital malformations and fetal death. CHIKV, despite not being classified as a preferentially neurotropic virus, causes encephalitis, meningitis and Guillain-Barré syndrome. Recently, the cultivation of cells in 3D systems was able to generate structures that mimic in vitro a microenvironment of cellular organization that occurs in vivo. In this context, human neuroblastoma cells, SH-SY5Y, represent a promising cell model for 3D cultivation. It is not fully understood how ZIKV and CHIKV act in neurogenesis, in which developmental regulatory genes are involved, such as the HOX genes (Homeobox), which participate in the differentiation of different types of cells. Thus, the general objective of the study was to develop new 3D neuronal culture models (neurospheres) from SH-SY5Y cells and to evaluate the modulation of HOX gene expression in these models in the process of cell differentiation after infection with ZIKV and CHIKV. For this, three-dimensional cultivation of SH-SY5Y cells was performed using the magnetic levitation technique. The neurospheres were differentiated into mature neurons with all trans retinoic acid and BDNF (Brain derived neurotrophic factor) and infected with ZIKV and/or CHIKV. The expression of the HOX genes was analyzed by RT-qPCR and by flow cytometry the inflammatory profile of the infected neurospheres. It was observed that the neurospheres are able to differentiate into neurons, are permissive to infection by both viruses and generate an inflammatory response with increased levels of IL-6 and IL-8 for both ZIKV and CHIKV. Infection with these viruses significantly alters the expression of genes SIX2, SIX3, SIX4 and SIX6 involved in the process of cell differentiation. Other HOX genes, such as ARX, HOXD1, DMBX1, POU5F1, had their expression significantly altered depending on the virus. These results suggest that neurospheres are viable models for the study of viral neuropathogenesis and the alteration of HOX gene expression reveals important aspects of these viruses in the course of neurogenesis.
