A resistência de larvas de Culex quinquefasciatus à toxina Binária (Bin) de Lysinibacillus sphaericus, pode ser causada por mutações no gene do receptor da toxina Bin. Em nosso laboratório, uma colônia resistente selecionada com o L. sphaericus cepa IAB59 vem sendo utilizada como modelo de estudo e sua refratariedade é causada pelo alelo cqm1REC que impede a expressão do receptor Cqm1. Um estudo prévio mostrou que as larvas resistentes, comparadas às suscetíveis, apresentam um transcriptoma diferencial com um padrão de transcrição alterado para genes ligados ao metabolismo de lipídios, carboidratos, proteínas, de xenobióticos, entre outros. O objetivo deste estudo foi validar funcionalmente e investigar estas moléculas que podem estar de alguma forma associadas ao status de resistência de Cx. quinquefasciatus. Para tal, a atividade de enzimas selecionadas foi determinada em intestinos de larvas resistentes e suscetíveis. Em relação às lipases, as larvas resistentes mostraram uma atividade menor para os substratos acetato (R= 331,5 ± 24,4, S= 381,8 ± 22,8), butirato (R= 206,3 ± 9,6, S= 257,7 ± 17,3) e heptanoato (R= 121,1 ± 10,06, S= 188,5 ± 14,8). A atividade sobre os substratos oleato, e palmitato não mostrou uma diferença significativa. A atividade de α-glicosidases utilizando o substrato sacarase mostrou uma atividade significativamente reduzida em larvas resistentes (R= 62,25 ± 4,8; S=112 ± 4), enquanto esta atividade foi similar para o substrato MuαGlu. A atividade de proteases e de enzimas detoxificadoras glutationa-S-transferases, por sua vez, foi similar para as duas colônias. Foram avaliadas as reservas de lipídios e açúcares redutores e o perfil de larvas e adultos da colônia resistente foi caracterizado pela redução significativa do acúmulo de lipídios (58%) e maior quantidade de açúcares redutores (35%). Contudo, a fertilidade e fecundidade foi semelhante para as duas linhagens, indicando que as alterações nas reservas não afetaram esses parâmetros. Este estudo também visou a identificação e caracterização parcial de uma panteteinase cujo transcrito foi o mais reprimido no transcriptoma de larvas resistentes. O gene foi clonado e expresso em células Sf9 e revelou uma proteína de cerca de 70 kDa, maior do que o predito de 57 kDa. A análise in silico da sequência apresentou três potenciais sítios de N-glicosilação. Uma discreta presença de N-glicosídeos (1 kDa) foi detectada em ensaios in vitro, porém este achado não justifica a discrepância entre os pesos, assim, a proteína deve sofrer outras alterações pós-traducionais. A quantificação relativa da transcrição de panteteinase em larvas em larvas suscetíveis mostrou uma grande variação na expressão entre indivíduos, enquanto o transcrito estava reprimido em todas as larvas resistentes (S= 2,3 ± 2, R= 0,19 ± 0,1). A investigação da expressão de panteteinase nativa em larvas mostrou uma proteína de 70 kDa que foi imunodetectada pelo anticorpo anti-panteteinase em amostras de microviili de intestino, o que sugere a sua localização como uma proteína de membrana. Em conclusão, Cx. quinquefasciatus resistentes à Bin possuem características metabólicas diferenciais e a identificação da panteteinase como um segundo marcador da resistência à toxina Bin abre perpectivas para investigação do papel destas moléculas.
Abstract
The resistance of Culex quinquefasciatus larvae to the Binary toxin (Bin) from Lysinibacillus sphaericus, can be caused by mutations in the receptor of this toxin. In our laboratory, a resistant colony selected with the L. sphaericus strain IAB59 has been used as a study model and its refractoriness is caused by the cqm1REC allele that prevents the expression of the Cqm1 receptor. A previous study showed that resistant larvae, compared to the susceptible ones, present a differential transcriptome with an altered transcription of genes linked to the metabolism of lipids, carbohydrates, proteins, xenobiotics, among others. The aim of this study was to obtain a functional validation and to investigate these molecules that may be somehow associated with'''the resistance status of Cx. quinquefasciatus. For this purpose, the activity of selected' enzymes was determined in midgut samples of resistant and susceptible larvae. In relation to lipases, the resistant larvae showed a lower activity for substrates acetate (R= 331,5 ± 24,4, S= 381,8 ± 22,8), butyrate (R= 206,3 ± 9.6, S= 257,7 ± 17,3) and heptanoate (R= 121,1 ± 10,06, S= 188,5 ± 14,8). The activity on oleate and palmitate substrates did not show a significant difference. The activity of α-glucosidases on sucrose showed a significantly reduced activity in resistant larvae (R= 62,25 ± 4,8; S=112 ± 4), while this activity was similar for the MuαGlu substrate. The activity of proteases and gluthatione-S-transferases detoxifying enzymes, in turn, was similar for both colonies. The reserves of lipids and reducing sugars were evaluated and the profile of larvae and adults of the resistant strain was characterized by a significant reduction in lipid accumulation (58%) and a higher amount of reducing sugars (35%). Fertility and fecundity were similar for both strains, indicating that changes in reserves did not affect these parameters. This study also aimed to identify the pantetheinase whose transcript was the most repressed in the transcriptome of resistant larvae. The gene was cloned and expressed in Sf9 cells and revealed a protein of about 70 kDa, different of the predicted molecular weight of 57 kDa. The in-silico analysis of the protein sequence showed three potential N-glycosylation sites. A slight presence of N-glycosides (~1 kDa) was detected by in vitro assays, but this finding does not justify the discrepancy between the predicted and real molecular weights, and it is likely that other post-translational modifications should be responsible for that. The relative quantification of the panthethein transcripts in susceptible larvae showed its expression characterized by a wide variation among individuals, while those transcripts 'were repressed in all resistant larvae (S= 2.3 ± 2, R= 0.19 ± 0.1). The investigation of 'the native pantetheinase in larvae showed a 70 kDa protein which was immunodetected by anti-pantheteinase antibodies in samples of midgut microvilli, suggesting its expression as a membrane-bound protein. In conclusion, Cx. quinquefasciatus Bin-resistant have differential metabolic characteristics and identification of the pantetheinase as a second marker of Bin-resistance opens perspectives for the investigation of the role played by these molecules.
