Os tripanossomatídeos patogênicos são responsáveis por causar doenças tropicais negligenciadas que afetam milhares de pessoas em escala mundial, motivando pesquisas para desenvolver terapias mais eficazes. A síntese de proteínas (tradução) é um alvo em potencial para quimioterápicos, sendo essencial em todos os organismos e regida por fatores de iniciação eucarióticos (eIFs). Nesse contexto, o eIF4A é uma RNA helicase da família DEAD-box que faz parte do complexo eIF4F, juntamente com as demais subunidades eIF4G e eIF4E, desempenhando um papel fundamental no recrutamento do mRNA para o início do processo de tradução, sendo passível de inibição. Múltiplos complexos do tipo eIF4F foram descritos em tripanossomatídeos, entre os quais se destacam aqueles baseados nos homólogos EIF4G3, EIF4G4, EIF4E3, EIF4E4 e EIF4AI. Este projeto teve como objetivo aprofundar o conhecimento sobre aspectos funcionais e estruturais do fator EIF4AI em Trypanosoma brucei (Tb) e Leishmania infantum (Li). Os dados obtidos indicam um perfil de interação espécie-específica entre os fatores EIF4AI-EIF4G3/4(MIF4G), nos tripanossomatídeos. A avaliação in silico de motivos conservados do EIF4AI (NHTV/DLYE/GSSRVL) e do seu parceiro EIF4G (R_LNK) presentes nos tripanossomatídeos reforçam a importância de cada motivo para manutenção do complexo. A análise de contados moleculares mostrou a interação do motivo R_LNK com o DLYE, um contato importante para a manutenção dos complexos EIF4AI-EIF4G, e mostrou um mecanismo diferencial de interação do complexo EIF4AI-EIF4G4 de L. infantum, caracterizado por um impedimento estérico em virtude de um resíduo de arginina (R) vizinha ao motivo (R_LNK). Outro ponto chave para a interação entre esses fatores é a arginina 39 (R39) de Tb/LiEIF4AI que interage com um motivo onde a fenilalanina (F) é bastante conservada no EIF4G3/4, sendo esta a maior contribuição para a manutenção do complexo nestes parasitas. Como parte da validação experimental dos motivos funcionais e de interação, a expressão da proteína TbEIF4AI em formas mutantes (SAT→AAA) e TbEIF4AI (DLYE→ALYA) reduzem a taxa de crescimento das linhagens de T. brucei. A análise do perfil de ligação ao inibidor específico de eIF4A, Rocaglamide A, e com o RNA (EIF4AI:RNA:RocA) apontou um perfil de interação diferencial entre T. brucei e L. infantum, e em comparação com humanos. Em T. brucei, a presença de uma valina (V160) no sítio de ligação gera interações mais fracas, reduzindo a estabilidade do RocA no seu sítio, enquanto em L. infantum, na mesma posição tem uma serina (S159) que permite o acesso de um resíduo de lisina (K163) ao sítio de ligação, garantido uma interação mais forte, e que estabiliza melhor o inibidor RocA no sítio de ligação .
Abstract
Pathogenic trypanosomatids are responsible for causing neglected tropical diseases that affect thousands of people worldwide, motivating research to develop more effective therapies. Protein synthesis (translation) is a potential target for chemotherapeutic drugs, being essential in all organisms and governed by eukaryotic initiation factors (eIFs). In this context, eIF4A is an RNA helicase from the DEAD-box family that is part of the eIF4F complex, together with the other subunits eIF4G and eIF4E, playing a fundamental role in recruiting mRNA for the beginning of the translation process, being subject to inhibition . Multiple eIF4F-type complexes have been described in trypanosomatids, among which those based on the homologues EIF4G3, EIF4G4, EIF4E3, EIF4E4 and EIF4AI stand out. This project aimed to deepen knowledge about functional and structural aspects of the EIF4AI factor in Trypanosoma brucei (Tb) and Leishmania infantum (Li). The data obtained indicate a species-specific interaction profile between the EIF4AI-EIF4G3/4(MIF4G) factors in trypanosomatids. The in silico evaluation of conserved motifs of EIF4AI (NHTV/DLYE/GSSRVL) and its partner EIF4G (R_LNK) present in trypanosomatids reinforce the importance of each motif for maintaining the complex. Analysis of molecular counts showed the interaction of the R_LNK motif with DLYE, an important contact for the maintenance of the EIF4AI-EIF4G complexes, and showed a differential mechanism of interaction of the L. infantum EIF4AI-EIF4G4 complex, characterized by a steric hindrance in due to an arginine residue (R) neighboring the motif (R_LNK). Another key point for the interaction between these factors is arginine 39 (R39) of Tb/LiEIF4AI, which interacts with a motif where phenylalanine (F) is quite conserved in EIF4G3/4, this being the greatest contribution to the maintenance of the complex in these parasites. As part of the experimental validation of the functional and interaction motifs, the expression of the TbEIF4AI protein in mutant forms (SAT→AAA) and TbEIF4AI (DLYE→ALYA) reduces the growth rate of T. brucei strains. Analysis of the binding profile to the specific eIF4A inhibitor, Rocaglamide A, and with RNA (EIF4AI:RNA:RocA) pointed to a differential interaction profile between T. brucei and L. infantum, and in comparison with humans. In T. brucei, the presence of a valine (V160) in the binding site generates weaker interactions, reducing the stability of RocA at its site, while in L. infantum, in the same position there is a serine (S159) that allows access of a lysine residue (K163) to the binding site, ensuring a stronger interaction, and better stabilizing the RocA inhibitor at the binding site .
