Nos óleos e gorduras usados em frituras são formadas substâncias relacionadas a um grande número de patologias. Os grupos dos monoepoxiácidos (EPOXI), monocetoácidos (CETO) e monohidroxiácidos (HIDROXI) produzidos na fritura são relevantes sob o ponto de vista quantitativo. A toxicidade dos EPOXI e de seus metabólitos tem sido demonstrada. O Brasil não possui legislação federal para uso destes óleos e gorduras e as legislações internacionais estabelecem parâmetros obtidos através de análises laboratoriais. Porém, os serviços de fiscalização nesta área requerem testes rápidos aplicáveis in situ. No entanto, não há teste rápido cuja confiabilidade tenha sido comprovada. Pelo exposto, para contribuir com estudos de avaliação de risco de alimentos fritos e com uma fiscalização efetiva e eficiente nesta área, os objetivos deste estudo foram obter dados de teor de EPOXI, CETO e HIDROXI e avaliar a citotoxicidade de monoepóxido formado significativamente; como também validar os testes rápidos. Os óleos refinados de palma e de soja e a gordura de soja parcialmente hidrogenada provenientes de experimentos de fritura descontínua de batatas foram analisados. Foram utilizados os testes rápidos Viscofrit (VF) e Food oil sensor (FOS) e os métodos analíticos de determinação de ácidos graxos, polímeros, EPOXI, CETO e HIDROXI, através de técnicas cromatográficas a líquido de partição por tamanho de partículas, de adsorção em fase sólida, e a gás de alta resolução. Também foi avaliada a citotoxicidade in vitro do ácido trans-9,10-epoxiesteárico (EPO), monoepoxiácido formado em quantidades relevantes a partir de óleos e gorduras oriundos de fritura de batatas, em células de hepatoma de ratos, HTC (Hepatoma Tissue Culture), através do ensaio de viabilidade celular mediado pelo diacetato de fluoresceína e brometo de etídio. Os resultados obtidos neste estudo demonstraram: que o óleo de palma e a gordura hidrogenada apresentam maior tendência à formação de EPOXI, CETO e HIDROXI, e menor tendência à degradação do que o óleo de soja. Os grupos mencionados acima constituem quantitativamente uma fração muito importante das substâncias de degradação. O teste rápido FOS foi o de escolha para decidir o momento de descarte de óleos e gorduras não conformes. O EPO foi citotóxico em células hepáticas HTC.
Abstract
Substances related to a high number of diseases can be present in used frying fats and oils. Among the new groups of compounds originated at frying temperatures, monoepoxyacids, monocetoacids and monohydroxyacids are quantitatively important. Toxicity of monoepoxyacids and its metabolites has been demonstrated. Brazil does not have Federal Laws regulating these oils and fats. However, international legislations in many countries have established parameters obtained from laboratorial analyses to limit the degradation of used frying fats and oils for human consumption. In this respect, inspections in this area require rapid tests to be applied in situ. There isn’t rapid test which reliability has been confirmed. Contributing with risk assessment of the frying foods and the effective inspection, the targets of this work were to obtain data of monoepoxyacids, monocetoacids and monohydroxyacids contents and evaluate the cytotoxicity of the significantly formed monoepoxyde, as well as validate the rapid tests. Soybean oil, palm oil and partially hydrogenated vegetable fat used in frying to prepare chips were analyzed. Rapid tests based on physical properties Viscofrit (VF) and Food oil sensor (FOS) were evaluated to determine the point at which frying fats and oils should be discarded. Also, chromatographic techniques based on molecular size distribution, solid phase adsorption and high-resolution gas were applied for quantitative determination of fatty acids, polar compounds, polymers, polar fatty acids, monoepoxyacids, monocetoacids and monohydroxyacids. Cytotoxicity in vitro of trans-9, 10-epoxystearic acid in HTC cells through out phluorescein di-acetate and ethidium bromide was also evaluated. The results showed that palm oil and partially hydrogenated vegetable fat present higher tendency to the formation of monoepoxyacids, monocetoacids and monohydroxyacids, than soybean oil. The groups mentioned above are very important from a quantitative point of view among the fatty acids undergoing degradation. Rapid test FOS was the choice to decide the moment of discarding used frying oils and fats according to the present international regulations. Trans-9, 10-epoxystearic acid was cytotoxic in hepatic HTC cells.
