As espécies do gênero Passiflora possuem muita utilização na medicina tradicional, pois apresentam diversas aplicações. Inúmeras espécies desse gênero possuem comprovação científica de suas atividades sedativas e ansiolíticas, pois apresentavam efeitos significativos no sistema nervoso central. A espécie que mais possui estudos a respeito da sua atividade farmacológica é a Passiflora incarnata (Pi). O objetivo deste trabalho foi avaliar a eficácia da Pi, de fabricantes distintos, quanto ao seu efeito ansiolítico através de testes neurocomportamentais e alterações morfofuncionais, além de realizar testes de controle de qualidade microbiológico e físico-químicos. Utilizou-se camundongos Swiss Webster, machos, separados em 5 grupos experimentais (Veículo PBS, Pi referência 100, 200, 400 e 800 mg/kg) para obtenção da curva de doses, e foram realizados os ensaios comparativos entre a Pi referência e outras 3 marcas comerciais, nas doses de 200 e 800 mg/kg. Um grupo tratado com um produto natural não ansiolítico (Castanha da Índia) foi adicionado aos testes como uma espécie de controle negativo. Os animais receberam 3 doses em diferentes momentos, 1 hora antes do teste do Labirinto em Cruz Elevado (LCE), 1 hora antes do teste Campo Vazado (CV) e 1 hora antes da eutanásia. Após a eutanásia, amostras de sangue e tecido cerebral foram recolhidas para análises bioquímicas e quantificação da proteína BDNF por ELISA. As amostras de medicamentos comerciais de Pi foram analisadas através de ensaios microbiológicos e físico-químicos (peso médio e cromatografia em camada delgada (CCD)) estabalecidos pela Farmacopeia Brasileira (FB). O tratamento com Pi referência nas doses de 400 e 800 mg/kg revelou um aumento significativo no tempo de permanência nos braços abertos no LCE, indicativo de uma atividade ansiolítica. No teste do CV, os animais não apresentaram diferença no número de deslocamentos, em contrapartida, os grupos de 400 e 800 mg/kg realizaram mais head dipping em comparação ao grupo PBS. Comparando as marcas comerciais com a Pi referência, foi possível observar um comportamento semelhante do grupo referência com as Marcas 1 e 3, apenas a Marca 2 não apresentou diferenças significativas, tanto no teste LCE como no CV. Observamos diferenças significativas nos parâmetros AST e Creatina Quinase entre os grupos experimentais na análise bioquímica, e a maioria dos valores encontrados estava dentro da faixa de valores padrões. Na quantificação de BDNF foi possível observar um resultado discrepante apenas para a Marca 1, indicando que a dose de 200 mg/kg nessa marca não é suficiente para tratar ansiedade. Observamos um perfil fitoquímico obtido após CCD semelhante em todas as amostras analisadas, indicativo de que todas as marcas apresentam as mesmas substâncias químicas que são responsáveis pela ação ansiolítica do fitoterápico. A análise do parâmetro físico de peso médio demonstrou que todas as marcas cumpriam com os limites estabelecidos pela FB. Nos ensaios microbiológicos observou-se crescimento bacteriano na Marca 2 (103 UFC/g), confirmada pela presença de Staphylococcus epidermidis. Observou-se o crescimento fúngico nas Marcas 2 e 3, (102 UFC/g), e após determinação por morfologia, detectou-se a presença de Aspergillus sp.. Os resultados encontrados estão dentro dos parâmetros estabelecidos pela FB.
Abstract
The species of the genus Passiflora are widely used in traditional medicine, as they have several applications. Numerous species of this genus have scientific evidence of their sedative and anxiolytic activities, as they have significant effects on the central nervous system. The species that has more studies about it is the Passiflora Incarnata (Pi). The objective of this work is to evaluate the effectiveness of Pi, from different manufacturers, regarding its anxiolytic effect through neurobehavioral tests and morphofunctional changes, in addition to carrying out microbiological and physical-chemical quality control tests. Male Swiss Webster mice were used, separated into 5 experimental groups (PBS vehicle, Pi reference 100, 200, 400 and 800 mg/kg) to obtain the dose curve, and then comparative tests between Pi reference and 3 other commercial brands, in doses of 200 and 800 mg/kg. A group treated with a non-anxiolytic natural product (Horse Chestnut) was added to the tests as a kind of negative control. The animals received 3 doses at different times, 1 hour before the Elevated Plus Maze (EPM) test, 1 hour before the Hole Board (HB) test and 1 hour before euthanasia. After euthanasia, blood and brain tissue samples were collected for biochemical analysis and BDNF protein quantification by ELISA. In addition, commercial Pi drug samples were analyzed using microbiological and physicochemical assays (mean weight and thin layer chromatography (TLC)) established by the Brazilian Pharmacopoeia (BP). Treatment with reference Pi at doses of 400 and 800 mg/kg revealed a significant increase in the time spent in the open arms in the EPM, indicative of anxiolytic activity. In the HB test, the animals showed no difference in the number of displacements, on the other hand, the 400 and 800 mg/kg groups performed more head dipping compared to the PBS group. Comparing the commercial brands with the Pi reference, it was possible to observe a similar behavior of the reference group with Brands 1 and 3, only Brand 2 did not show significant differences, both in the EPM test and in the HB. Again, there was no difference in the number of displacements, however, all 800 mg/kg groups, with the exception of Brand 2, performed more head dipping compared to the PBS group. We observed few significant differences between the experimental groups in the biochemical analysis, and most of the values found were within the range of standard values. In the quantification of BDNF, it was possible to observe a discrepant result only for Brand 1, indicating that the dose of 200 mg/kg in this brand is not enough to treat anxiety. We observed a very similar phytochemical profile obtained after TLC in all analyzed samples, indicating that all brands present the same chemical substances that are responsible for the anxiolytic action of the herbal medicine. The analysis of the physical parameter of average weight showed that all brands complied with the limits established by BP. In the microbiological assays, bacterial growth was observed at Mark 2 (103 CFU/g), confirmed by the presence of Staphylococcus epidermidis. Added to this, fungal growth was observed in Brands 2 and 3, (102 CFU/g), and after determination by morphology, the presence of Aspergillus sp. was detected. The results found are within the parameters established by BP.
