A dengue é uma das principais arboviroses humana, causada por um dosquatro sorotipos virais (DENV-1, DENV-2, DENV-3 e DENV-4). A infecção porqualquer um dos quatro sorotipos pode resultar desde uma forma clínicainaparente a manifestações clínicas graves. O genoma viral consiste de RNA,que após tradução e processamento, origina dez proteínas: três estruturais esete não estruturais (NS), envolvidas na replicação viral. Estudos buscamcorrelacionar fatores genéticos do hospedeiro com a predisposição àresistência, suscetibilidade ou gravidade da infecção pelo dengue. A respostaimune 72 de pacientes com dengue foi analisada ex-vivo, utilizando PeripheralBlood Mononuclear Cells (PBMC) de pacientes provenientes de uma coorte dedengue previamente iniciada no Departamento de Virologia e TerapiaExperimental (LAVITE/CPqAM). Essa análise foi realizada através de EnzymeLinked Immunospot (ELISPOT), utilizando-se peptídeos sintéticos abrangendoas proteínas NS1 e NS3 do DENV-3, para estimular células T CD4+ e CD8+. Asrespostas imunes foram analisadas quanto à associação com atributos clínicose imunogenéticos, tais quais: forma clínica, tipo de infecção e HumanLeukocyte Antigen (HLA) dos pacientes e seus supertipos. A identificação ex-vivo de epítopos para células T obtidas experimentalmente também foicomparada com as previsões teóricas utilizando-se algoritmos computacionaisavançados. Os resultados evidenciaram que seis peptídeos, sendo três da NS1e três da NS3, foram propostos como epítopos de células T. Em acréscimo, opeptídeo NS1_297_311 apresentou perfil de positividade alélica entre apopulação e o gene HLA-DRB1. Esses achados podem contribuir para odesenvolvimento de intervenções terapêuticas ou preventivas, como umavacina baseada em epítopos múltiplos contra a dengue.
Abstract
Dengue is the most important mosquito-borne viral disease caused by one offour serotypes (DENV-1, DENV-2, DENV-3 and DENV-4). Infection by any ofthe four serotypes may result from a clinical form with no symptoms to severeclinical manifestations. The viral genome consists of RNA, which aftertranslation and processing, will result in ten proteins: three structural and sevennonstructural (NS), which are involved in viral replication. Studies seek tocorrelate with genetic factors host predisposing to resistance, susceptibility orseverity of dengue infection. The immune response of dengue patients (n = 72)was analyzed ex vivo, using Peripheral Blood Mononuclear Cells (PBMC) ofpatients from a cohort of dengue previously started at the Department ofVirology and Experimental Therapy (LaViTE/ CPqAM). This analysis wasperformed by Enzyme Linked Immunospot (ELISPOT), using synthetic peptidesspanning the proteins NS1 and NS3 of DENV-3, to stimulate CD4+ and CD8+ Tcells. Immune responses were analyzed for association with clinical andimmunogenetic attributes, such as: clinical form, type of infection and theHuman Leukocyte Antigen (HLA) of the patient and their supertypes. Theidentification of ex-vivo T-cell epitopes obtained experimentally was alsocompared with theoretical predictions using advanced computationalalgorithms. Our results showed that six peptides, three of NS1 and three of NS3proteins, were proposed as T cell epitopes. In addition, the peptideNS1_297_311 showed profile allele positivity among the population and HLA-DRB1 gene. These findings may contribute to the development of preventive ortherapeutic interventions, such as a multi-epitope-based vaccine againstdengue.
