Atualmente, são complexas as ações que envolvem as práticas de Vigilância Sanitária, necessitando de políticas públicas estabelecidas pelo Sistema Único de Saúde (SUS) de promoção, prevenção e proteção da saúde. O Instituto Nacional de Controle de Qualidade em Saúde (INCQS) tem como objetivo contribuir para a promoção e recuperação da saúde e prevenção de doenças, além de atuar como referência nacional para as questões científicas e tecnológicas relativas ao controle da qualidade de produtos, ambientes e serviços no âmbito da Vigilância Sanitária. O Setor de Cultura de Células (SCC) do INCQS participa dos processos de manutenção e fornecimento do acervo de linhagens celulares animais. Essas linhagens são amplamente empregados no controle da qualidade de cosméticos e imunobiológicos, em isolamento e pesquisa de vírus, bem como em métodos alternativos em substituição ao uso de animais. Logo é fundamental avaliar a conformidade de todo material biológico utilizado. A contagem de células viáveis de células eucarióticas é o primeiro passo na manutenção e expansão de culturas celulares in vitro, sendo importante para o monitoramento rotineiro da saúde celular e da taxa de proliferação, e para a semeadura de células para uso em experimentos subsequentes. Existem vários corantes utilizados na contagem das células, o mais usado é o Azul de Tripan, porém já foi constatado que apresenta efeito teratogênico. A Eritrosina B é utilizada principalmente como corante alimentar, porém já foi demonstrado que pode servir como meio alternativo para a contagem de células. Como a biossegurança no INCQS é uma preocupação crescente, o uso da Eristrosina B é uma opção para diminuir o uso do Azul de Tripan. O objetivo do trabalho é realizar um estudo comparativo com o corante Azul de Tripan e a Eritrosina B por meio das contagens das linhagens celulares VERO e L-929 em câmara de Neubauer. Foi realizado o cultivo celular das linhagens VERO segundo o POP 65.3430.031 e NCTC 929 segundo o PU.3430.055. A contagem e a verificação da viabilidade das células foram feitas utilizando os corantes Azul de Tripan e a Eritrosina B seguindo o PU.3430.053. A análise estatística foi realizada para avaliar a normalidade dos resultados e constatando a não normalidade, foi aplicado um teste não paramétrico. A partir da análise estatística, contatamos que o teste de equivalência referente a comparação das médias das concentrações da célula VERO e L-929 com os dois corantes não detectou diferenças estatísticas significativas. Porém, a comparação das médias das viabilidades da célula VERO e L-929 com os dois corantes houve diferença estatística significativa. Apesar de a Eritrosina B não ser estatisticamente similar ao Azul de Tripan, pode vir a ser utilizado como corante de cultura celular, desde que seja estabelecido um método confiável para que se obtenha um resultado fidedigno de concentração e viabilidade.
Abstract
Currently, actions involving Health Surveillance practices are complex, requiring public policies established by the Unified Health System (SUS) for health promotion, prevention and protection. The National Institute for Health Quality Control (INCQS) aims to contribute to the promotion and recovery of health and disease prevention, in addition to acting as a national reference for scientific and technological issues relating to quality control of products, environments and services within the scope of Health Surveillance. The INCQS Cell Culture Sector (SCC) participates in the maintenance and supply processes of the collection of animal cell lines. These strains are widely used in the quality control of cosmetics and immunobiologicals, in virus isolation and research, as well as in alternative methods to replace the use of animals. Therefore, it is essential to evaluate the conformity of all biological material used. Counting viable cells of eukaryotic cells is the first step in maintaining and expanding in vitro cell cultures and is important for routine monitoring of cellular health and proliferation rate, and for seeding cells for use in subsequent experiments. There are several dyes used in cell counting, the most used is Trypan Blue, but it has been found to have a teratogenic effect. Erythrosine B is mainly used as a food coloring, but it has been shown that it can serve as an alternative means for cell counting. As biosafety at INCQS is a growing concern, the use of Erythrosin B is an option to reduce the use of Trypan Blue. The objective of the work is to carry out a comparative study with the dye Trypan Blue and Erythrosin B by counting VERO and L-929 cell lines in a Neubauer chamber. Cell culture of VERO lines was carried out as described in POP 65.3430.031 and NCTC 929 as described in PU.3430.055. The counting and verification of cell viability were carried out using the dyes Trypan Blue and Erythrosin B following PU.3430.053. Statistical analysis was performed to evaluate the fit of data to the normal distribution and, upon confirming non-normality, a non-parametric test was applied. From the statistical analysis, we found that the equivalence test referring to the comparison of the mean concentrations of the VERO and L-929 cells with the two dyes no statistically significant differences. However, the comparison of the mean viability of VERO and L-929 cells with the two dyes demonstrated statistically significant differences. Although Erythrosin B is not statistically similar to Trypan Blue, it can be used as a cell culture dye, as long as a reliable method is established to obtain a reliable concentration and viability result.
