A crescente tendência mundial pela substituição do uso de animais em experimentação promoveu a busca por métodos alternativos. Particular ênfase é dada ao desenvolvimento de métodos para substituir o tradicional Teste de Draize, utilizado para acessar a irritação ocular. Neste estudo, o ensaio de quantificação de proteínas totais em células SIRC e 3T3 utilizando o corante Azul Brilhante de Coomassie R-250 foi avaliado quanto ao seu valor em predizer o potencial de irritação ocular de cinco tensoativos. A concentração que induziu 50% de inibição do crescimento celular relativa aos controles (IC50) foi calculada para cada substância-teste. O modelo de citotoxicidade proposto apresentou boa capacidade de diferenciar tensoativos quanto ao seu potencial irritante, sobretudo quando conduzido com células SIRC. Apesar da necessidade de se testar um maior número de amostras e da realização de estudos inter-laboratoriais, esses resultados preliminares indicam que o ensaio de quantificação de proteínas totais com corante Azul Brilhante de Coomassie R-250, além de ser um método com características importantes (rapidez, sensibilidade, simplicidade de execução, baixo custo e alto grau de automação), é capaz de predizer o potencial de irritação ocular de tensoativos.
Abstract
The growing world tendency for the animal replacement in experimentation promoted the search for alternative methods. Particular emphasis is given to developing methods to replace the traditional Draize eye irritation test. In this study, the total protein cytotoxicity assay using SIRC and 3T3 cells and
the Coomassie Brilliant Blue R-250 dye was evaluated regarding its applicability for predicting ocular irritancy of surfactants. Our data show that the proposed cytotoxicity model has good ability to differentiate surfactants in relation to their irritancy potential, particularly when it was conducted with SIRC cells. Despite the need to test a greater number of samples and carrying out inter-laboratory
studies, these preliminary results indicate that the total protein cytotoxicity assay using Coomassie Brilliant Blue R-250 is a method with important characteristics (speed, sensibility, simplicity of implementation, low cost and high degree of automation) and is able to predict the ocular irritation potential of surfactants.
