Abstract in Portuguese | No Brasil é restrita a oferta de ensaios de proficiência (EP) na área de microbiologia de alimentos. Além disso, os altos custos diminuem a participação regular de laboratórios públicos e privados nesses ensaios. É de grande importância o incentivo da participação de laboratórios brasileiros em EP. Os materiais de referência (MR)utilizados para essa proposta devem apresentar características de homogeneidade e estabilidade que assegurem a distribuição de unidades com propriedades semelhantes, próximas a um valor padrão. A produção de MR destinados a ensaios microbiológicos é dificultada pela instabilidade natural dos microrganismos. O presente estudo avaliou a técnica de liofilização no preparo de MR para EP nos ensaios de detecção de Salmonella sp. e enumeração de B. cereus em leite. Foram preparados lotes de materiais contendo apenas um microrganismo e lotes mistos contendo Salmonella sp. e B. cereus em uma mesma matriz. Foram determinadas as concentrações de inóculo utilizadas no preparo dos diferentes lotes de MR e padronizados os procedimentos de contaminação da matriz. Para monitorar a qualidade dos materiais produzidos foram estabelecidos ensaios de controle, dentre eles o teste da homogeneidade e da estabilidade em longo e curto prazo. A técnica de liofilização se mostrou adequada como processo de dessecação para os MR produzidos. Os microrganismos se mantiveram estáveis à temperatura de estoque de -20ºC, no entanto, apresentaram perdas de viabilidade, quando estocados a temperaturas de 25ºC e 35ºC. Das bactérias estudadas B. cereus apresentou, em média, maior resistência às condições de dessecação. A padronização da técnica de produção desses MR propiciará a geração de materiais na área de microbiologia de alimentos, que poderão ser utilizados em futuros EP, com custos de participação mais acessíveis aos laboratórios nacionais. | pt_BR |
